银杏黄酮对脑缺血再灌注损伤大鼠脑组织的保护作用 被引量:6

The protective effect of ginkgo on ischemic-reperfusion brain injury in rats

摘要 目的探讨银杏黄酮对脑缺血再灌注损伤大鼠脑组织的保护作用及其机制。方法制备脑缺血再灌注脑损伤大鼠模型,并随机分为假手术组、模型组和银杏黄酮组,观察大鼠日常行为,并对实验大鼠脑进行病理组织学观察,检测脑组织匀浆中还原性谷胱甘肽(GSH)水平及过氧化氢酶(CAT)活力,Western blot检测大鼠脑组织中NF-κB蛋白表达水平。结果模型组大鼠活动减少、部分大鼠出现明显的神经缺损症状;模型组大鼠脑组织水肿,皮质神经细胞层次紊乱;银杏黄酮组与模型组比较,病理改变减轻;模型组神经细胞基质密度减低,胞质内空泡形成,未见明显核仁结构;银杏黄酮组神经细胞变性程度较模型组缓解,细胞核内染色质较均匀;与模型组比较,银杏黄酮组的GSH水平及CAT活力均升高(P<0.05);Western blot检测显示,模型组NF-κB蛋白水平明显升高,与假手术组比较有明显差异(P<0.05);银杏黄酮治疗后NF-κB蛋白水平降低,与模型组比较有明显差异(P<0.05)。结论银杏黄酮对缺血再灌注损伤大鼠脑组织有保护作用。 Objective To investigate the protective effect and mechanism of ginkgo on ischemic-reperfusion brain injury in rats. Methods Ischemic-reperfusion brain injury rat model were constructed and were randomly divided into control group, the model group and ginkgo group, daily behavior was observed, and the histological observation of experimental rats were done, glutathione(GSH) content and catalase(CAT) activity in brain tissue were detected, protein expression levels of NF-κB were also detected by western blot test. Results The animal activity in model group decreased, some animals appeared obvious neurologic symptoms. Brain tissue in control group were edema, nerve cells in cortex leveled disorder and loosely arranged, the pathological changes in ginkgo group reduced compared with control group. Cell matrix density in model group decreased, cytoplasmic vacuolation, there was no obvious nucleolus structure. Degree of nerve degeneration in ginkgo group relieved than that in model group, the nucleus chromatin were uniform. Compared with model group, GSH content and CAT activity in ginkgo group were higher(P<0.05).Western blot test results showed that the protein level of NF-κB in the model group increased significantly, compared with the control group(P<0.05), after treatment with ginkgo, the protein level of NF-κB decreased, the difference was statistically significant compared with the model group(P<0.05). Conclusion Ginkgo had protective effect in ischemic-reperfusion brain injury rats.
