山楂破壁饮片的HPLC指纹图谱研究

摘要 目的:建立山楂破壁饮片的高效液相(HPLC)指纹图谱,分析破壁饮片成品与中间体破壁粉及原料的化学相关性,为其整体质量评价提供依据。方法:采用C18柱,流动相为甲醇-乙腈-0.1%甲酸溶液,梯度洗脱流速为0.7mL/min,检测波长为280nm,柱温为30℃。结果:建立了以11个色谱峰为共有峰的山楂破壁饮片HPLC指纹图谱共有模式,指认了绿原酸、金丝桃苷2个共有峰;山楂破壁饮片与其破壁粉体(中间品)、传统饮片(原料)的图谱基本一致,相似度大于0.99。结论:建立的方法稳定、可靠、重复性好,可用于山楂破壁饮片整体质量分析。 Objective:To establish the HPLC fingerprint of the Ultrafine Granular Powder of Crataegi Fructus,and analyze the chemical composition relativity between the Ultrafine Granular Powder(UGP),Ultrafine Powder(UP)and its decoction pieces(DP),helping to evaluate its quality.Methods:RP-HPLC method was performed on an C18 column with a gradient elution composed of methanol-acetonitrile-0.1%formic acid at the flow rate of 0.7mL·min-1.The column temperature was set at 30℃,while the detective wavelength was set at 280nm.The fingerprint data analysis was carried out by using The Similarity Evaluation System of Chromatographic Fingerprint of Traditional Chinese Medicine of the National Pharmacopoeia Committee(2012).Results:The chromatographic fingerprint common pattern was established including 11 common characteristic peaks,and the Chlorogenic acid and Hyperoside were identified.The HPLC fingerprints were more than 0.99 similarity measurement between the UGP,UP and DP.Conclusion:The method with good reproducibility is reliable and stable,which is feasible for quality control of the Ultrafine Granular Powder of Crataegi Fructus.
