摘要
为石斛属种质资源保护与开发及新品种选育提供理论指导,采用ISSR分子标记技术对采自西南地区的23份石斛属植物和1份金石斛属植物资源进行遗传多样性和亲缘关系分析,并采用非加权平均距离法(UPGMA)进行聚类分析。结果表明:从20条ISSR标记引物中筛选出11条扩增条带清晰的引物,共检测出151个扩增位点,平均每条引物扩增位点为13.7个;其中多态性位点148个,多态性条带比率为98.01%;24份石斛总扩增条带为1084条,平均45.17条,其中多态性条带有1012条,占比为93.4%;24份石斛材料遗传多样性指数为0.63~0.78;在相似系数0.68处将24份石斛材料分为7个类群。西南地区石斛资源的遗传多样性非常丰富。
To provide theoretical guidance for the protection and development of Dendrobium germplasm resources and breeding of new varieties,the ISSR molecular marker technology was employed to analyze genetic diversity and relationship of 23 Dendrobium plants and one Flickingeria albopurpurea plant resources collected from southwestern China,and cluster analysis was also performed using the unweighted average distance method(UPGMA).Results:11 primers with clear amplification bands were selected from 20 ISSR-labeled primers,and a total of 151 amplification sites were detected,with an average of 13.7 amplification sites per primer;among them,polymorphic sites 148,with a polymorphic band ratio of 98.01%;The total number of amplified bands was 1084,with an average of 45.17,of which 1012 were polymorphic bands,accounting for 93.4%;The genetic diversity index was 0.63-0.78;at the similarity coefficient of 0.68,the 24 Dendrobium materials can be divided into seven groups.The genetic diversity of Dendrobium resources in Southwestern China was very rich.
