Establishment of a Deep Second Degree Scald Model in Rats and Detection of Molecular Markers

摘要 目的研究SD大鼠深Ⅱ度烫伤模型愈合的病理过程;探讨细胞角蛋白19(CK19)、血管内皮生长因子(VEGF)、胶原Ⅰ和胶原Ⅲ在愈合过程中的表达情况。方法采用自制的烫伤装置构建深Ⅱ度烫伤模型;应用组织HE切片进行病理学分析;采用逆转录PCR(RT-PCR)技术检测皮肤组织中CK19、VEGF、胶原Ⅰ和胶原Ⅲ在愈合过程中的表达情况。结果成功建立SD大鼠深Ⅱ度烫伤模型;创面组织CK19和胶原Ⅰ随着修复过程表达升高;VEGF先升高达峰值后再减低;胶原Ⅲ表达逐渐降低。结论创面表皮的修复是由附属结构中残存的上皮细胞分裂增殖完成。CK19、VEGF、胶原Ⅰ和胶原Ⅲ在修复过程中的表达呈明显变化,可作为烫伤修复的分子指标。 Objective To study the pathological process of deep second degree scald model healing in SD rats;to investigate the expression of cytokeratin 19(CK19),vascular endothelial growth factor(VEGF),collagen Ⅰ and collagen Ⅲ during healing.Methods A deep scald model was established by self-made scald device.Histopathological analysis was performed using tissue HE slices.The reverse transcription-PCR(RT-PCR)was used to detect expression situation of CK19,VEGF,collagen Ⅰ and collagen Ⅲ in skin tissue during healing.Results The deep second degree scald model of SD rats was successfully established.The expression of CK19 and collagen Ⅰ in wound tissue increased with the repair process;VEGF increased first and then decreased,and collagen Ⅲ decreased gradually.Conclusion The repair of wound epidermis is completed by the proliferation and proliferation of epithelial cells remaining in the accessory structure.The expression of CK19,VEGF,collagen Ⅰ and collagen Ⅲ showed significant changes during the repair process and could be used as a molecular marker for scald repair.
