摘要
目的:观察夹脊电针干预对大鼠脊髓损伤(SCI)区巨噬细胞NOGO受体(NgR)表达的影响。方法:Wistar大鼠48只,随机分为假手术组、模型组和2 Hz夹脊电针组、100 Hz夹脊电针组,每组12只。采用脊髓全横断造模方法造模,采用大鼠BBB运动功能评分于造模3、7、14 d评价脊髓损伤大鼠肢体运动功能,Western blot法测定各组巨噬细胞吞噬MBP的含量,比较各组巨噬细胞的吞噬能力。结果:与假手术组比较,各组大鼠BBB评分显著降低(P<0.05);与模型组比较,夹脊电针组BBB评分显著升高(P<0.05);与2 Hz夹脊电针组比较,100 Hz夹脊电针组BBB评分显著升高(P<0.05)。与模型组比较,夹脊电针组中表达NgR的巨噬细胞吞噬MBP显著增多(P<0.05);与2 Hz夹脊电针组比较,100 Hz夹脊电针组中表达NgR的巨噬细胞吞噬MBP显著增多(P<0.05)。结论:夹脊电针干预可明显增加脊髓损伤区巨噬细胞NgR的表达,从而增强巨噬细胞的吞噬能力,改善脊髓损伤大鼠肢体运动功能。
Objective:To investigate the effect of Jiaji electroacupuncture on NgR expression of macrophage in rats’spinal cord injury region.Methods:48 Wistar rats were randomly divided into sham operation group,model group)and 2 Hz Jiaji electroacupuncture group,100 Hz Jiaji electroacupuncture group with each 12 rats.Models were performed using a full-transverse modeling method of the spinal cord.The motor function of rats with spinal cord injury was observed by BBB scores after modeling 3d,7d and 14d.The content of MBP in macrophages of each group was determined by Western blot,the phagocytic ability of macrophages in each group was compared.Results:Compared with the sham operation group,the BBB scores of the other group rats were significantly lower(P<0.05),compared with the model group,the BBB score of the Jiaji electroacupuncture group was significantly increased(P<0.05);and the 2Hz Jiaji electroacupuncture group in comparison,the BBB score of the 100 Hz Jiaji electroacupuncture group was significantly increased(P<0.05).Compared with the model group,the phagocytosis of MBP by NgR-expressing macrophages in the Jiaji electroacupuncture groups were significantly increased(P<0.05);compared with the 2Hz Jiaji electroacupuncture group,the phagocytosis of MBP by NgR-expressing macrophagesiin the 100 Hz Jiaji electroacupuncture group were increased significantly(P<0.05).Conclusion:Jiaji electroacupuncture intervene can enhance the NgR expression of macrophage in rats’spinal cord injury region,enhance the phagocytic ability of macrophages and improve the limb motor function of rats with spinal cord injury.
