摘要
目的 探讨血清微小RNA-155(miR-155)联合组织人第10号染色体缺失的磷酸酶及张力蛋白同源的基因(PTEN)检测对子宫内膜癌的诊断价值。方法 选取2015年9月至2018年6月邯郸市第一医院妇产科收治的子宫内膜癌51例、良性病变患者52例,采用荧光实时定量聚合酶链反应(Q-PCR)法检测研究对象血清中miR-155表达,免疫组织化学法检测各组子宫内膜组织中PTEN水平,并通过受试者工作特征(ROC)曲线评价miR-155、PTEN及二者联合对子宫内膜癌的诊断价值。结果 与良性病变组相比,子宫内膜癌组血清miR-155表达水平显著升高,差异有统计学意义(P<0.05);与良性病变组相比,子宫内膜癌组PTEN蛋白表达量显著降低,差异有统计学意义(P<0.05);ROC曲线显示,血清miR-155水平预测患者子宫内膜癌发生的曲线下面积为0.876,灵敏度为96.3%,特异度为94.2%;子宫内组织PTEN水平预测患者子宫内膜癌发生的曲线下面积为0.952,灵敏度为94.1%,特异性度为92.3%;二者联合检测诊断子宫内膜癌曲线下面积为0.991,灵敏度为99.2%,特异度为92.5%。结论 血清miR-155联合组织PTEN检测可作为子宫内膜癌的诊断指标。
Objective To explore the diagnostic value of serum microRNA-155(miR-155)combined with tissue phosphatase and tensin homologous genes deleted on chromosome 10(PTEN)detection in endometrial cancer.Methods From September 2015 to June 2018,51 cases of endometrial cancer and 52 cases of benign lesions were selected,the expression of miR-155 in serum was detected by real-time fluorescence quantitative polymerase chain reaction(Q-PCR),PTEN level in endometrial tissues was detected by immunohistochemistry,the diagnostic values of miR-155,PTEN,and their combination in endometrial cancer were evaluated by receiver operating characteristic(ROC)curve.Results Compared with benign lesion group,the expression of serum miR-155 in endometrial cancer group was significantly increased,and the difference was statistically significant(P<0.05).Compared with benign lesion group,the expression of PTEN protein in endometrial cancer group was significantly decreased,and the difference was statistically significant(P<0.05).The ROC curve showed that the area under the curve of miR-155 in predicting the occurrence of endometrial cancer was 0.876,the sensitivity was 96.3%,and the specificity was 94.2%.The area under the curve of PTEN level in predicting the occurrence of endometrial cancer was 0.952,the sensitivity was 94.1%,and the specificity was 92.3%.The area under the curve of combined detection was 0.991,the sensitivity was 99.2%,and the specificity was 92.5%.Conclusion Serum miR-155 combined with tissue PTEN detection can be used as a diagnostic index for endometrial cancer.
