大鼠颅脑损伤联合FK506诱导促进坐骨神经再生的研究

摘要 目的:通过建立动物模型,比较大鼠脑损伤后联合FK506干预损伤神经的愈合情况,推测脑损伤合并坐骨神经损伤后应用FK506干预的可行性。方法:动物实验造模采用颅脑损伤模型(Feeney法)和坐骨神经损伤模型(Sunderland V型),实验组(A1组、A2组):颅脑损伤+坐骨神经损伤,对照组(B1组、B2组):单纯坐骨神经损伤,分为四组,第4、8、12周,进行测定坐骨神经指数,造模后8周、12周观察动作电位恢复率,造模后12周观察脊髓运动神经元荧光金的逆行示踪标记。结果:颅脑损伤联合应用FK506大鼠(A1组)周围神经损伤的修复效果优于其他组(A2组、B1组、B2组)。结论:大鼠脑损伤联合应用FK506干预可促进坐骨神经损伤修复,颅脑损伤与FK506促进神经修复作用的机制不完全相同,脑损伤促进周围神经损伤修复的机制仍需进一步深入研究。 Objective To compare the healing of injured nerves with FK506 after brain injury in rats by establishing an animal model experiment,and to speculate the feasibility of using FK506 after brain injury combined with sciatic nerve injury.Method Animal model was established by brain injury model(Feeney method)and sciatic nerve injury model(Sunderland V type),experimental group(A1,A2):craniocerebral injury+sciatic nerve injury,control group(B1,B2):simple sciatic nerve injury The rats were divided into 4 groups.The 4 th,8 th,and 12 th week were used to measure the sciatic nerve index.The recovery rate of action potential was observed at 8 and 12 weeks after modeling.The retrograde tracer of fluorescent gold in spinal motor neurons was observed 12 weeks after modeling.Results The repair effect of brain injury combined with FK506 rat(A1)peripheral nerve injury was better than other groups(A2,B1,B2).Conclusion Rat brain injury combined with FK506 intervention can promote the repair of sciatic nerve injury.The mechanism of brain injury and FK506 promoting nerve repair is not exactly the same.The mechanism of brain damage promoting peripheral nerve injury repair needs further study.
