摘要
目的观察乳腺小叶增生筛选方外用对乳腺增生模型大鼠乳腺组织病理及性激素受体的影响。方法将60只大鼠随机分为对照组、模型组、治疗组,每组20只。模型组及治疗组大鼠通过苯甲酸雌二醇序贯黄体酮肌肉注射复制乳腺增生模型,从肌肉注射开始,对照组及模型组给予淀粉糊精贴敷,治疗组给予乳腺小叶增生筛选方药糊贴敷,每日贴敷8 h以上,连续15 d。分别于干预5 d和10 d观察各组大鼠乳腺组织病理学表现,通过Image-pro-Plus 6.0软件分析各组大鼠腺泡面积及导管内径,免疫组化法检测各组大鼠雌激素受体(ER)、孕激素受体(PR)表达情况。结果治疗组大鼠干预5 d、10 d后,镜下见个别腺泡有轻度增生,无扩张。模型组和治疗组干预5 d和10 d后大鼠乳腺腺泡面积和导管内径均明显大于同期对照组(P均<0.05),但治疗组的腺泡面积均明显小于同期模型组(P均<0.05),治疗组的导管内径与模型组比较差异均无统计学意义(P均>0.05)。模型组及治疗组干预5 d后的ER评分均明显高于对照组(P均<0.05),但治疗组明显低于模型组(P<0.05);模型组干预10 d后ER评分仍明显高于同期对照组(P<0.05),治疗组恢复至对照组水平(P>0.05);各组干预5 d和干预10 d后PR评分比较差异均无统计学意义(P均>0.05)。结论外源性性激素可以使大鼠乳腺腺泡面积及导管内径增大,小叶增生筛选方外用可以缩小乳腺增生大鼠乳腺腺泡面积,抑制乳腺组织ER阳性表达,但对PR表达无明显影响。
Objective It is to observe the effect of external application of breast lobular hyperplasia screening decoction on the pathology of breast tissue and sex hormone receptors in rats with hyperplasia of breast.Methods Sixty rats were randomly divided into control group,model group and treatment group,with 20 rats in each group.The mammary gland hyperplasia models were established by sequential intramuscular injection of progesterone benzoate in the rats in the model group and the treatment group.The control group and the model group were given amylodextrin by application,and the treatment group was given breast lobular hyperplasia screening decoction by application starting from the intramuscular injection.The application lasted more than 8 h daily for 15 d.The histopathological manifestations of mammary glands of rats in each group after 5 and 10 days of intervention were observed,and the acinar area and inner diameter of the catheter in each group was analyzed by Image-pro-Plus 6.0 software,and the expression of estrogen receptor(ER),progesterone receptor(PR)in each group was detected by immunohistochemistry.Results After 5 d and 10 d of intervention,slight hyperplasia without dilatation of individual acinars could be found under the microscope in the treatment group.After 5 and 10 days of intervention,the breast acinar area and inner diameter of the ducts of the rats in the model group and the treatment group were significantly larger than those in the control group(P<0.05),but the acinar area inthe treatment group was significantly smaller than that in the model group(P<0.05),there was no significant difference in the inner diameter of the catheter between the treatment group and the model group(all P>0.05).The ER scores in the model group and the treatment group after 5 days of intervention were significantly higher than those in the control group(both P<0.05),but the scores in the treatment group was significantly lower than that in the model group(P<0.05).The ER scores in the model group was still significantly higher than that in the control group after 10 days of intervention(P<0.05),the scores in treatment group returned to the same level as the control group(P>0.05);there was no significant difference in PR scores between every group after 5 days and 10 days of intervention(P>0.05).Conclusion Exogenous sex hormones can increase the area of breast acinus and the inner diameter of ducts in rats.The external use of breast lobular hyperplasia screening decoction can reduce the area of breast acinus in breast hyperplasia rats and inhibit the positive expression of ER in breast tissues,but it has no significant effect on PR expression.
