响应面法优化广西甜茶总黄酮提取工艺及抗氧化活性研究 被引量:6

Optimization of extraction technology and antioxidant activity of flavonodis from Rubus suavissimus S. by response surface methodology

摘要 广西甜茶是集茶、糖、药三位为一体的广西特有甜味植物,通过乙醇-超声波提取研究了料液比、提取温度、提取时间等条件对其总黄酮含量的影响,结合响应面分析得到了总黄酮的最优提取工艺,并测定了其抗氧化活性。结果表明,广西甜茶总黄酮最佳提取条件为料液比1:41 g/mL、超声功率340.8 W、提取温度73.5℃、提取时间101.4 min,总黄酮提取率为8.15%。抗氧化活性试验结果表明,黄酮浓度为2 mg/mL时,对·OH、DPPH·和O2^-·的最大清除率分别为70.1%、89.7%和80.6%,具有较好的活性。以期为广西甜茶产业普及化及开发新天然抗氧化剂提供理论依据。 Rubus suavissimus S. is a unique sweet plant in Guangxi, which integrates tea, sugar and medicine. The effects of material liquid ratio, extraction temperature and extraction time on the content of total flavonoids were studied by ethanol ultrasonic extraction. The optimal extraction technology of total flavonoids was obtained by response surface analysis, and its antioxidant activity was determined. The results showed that the best extraction conditions of total flavonoids were as follows: the ratio of material to liquid was 1:41 g/mL, the ultrasonic power was 340.8 W, the extraction temperature was 73.5 ℃, the extraction time was 101.4 min, and the extraction rate of total flavonoids was 8.15%. The results of antioxidant activity test showed that when the concentration of flavone was 2 mg/mL, the maximum scavenging rates of ·OH, DPPH· and O2^-· were 70.1%, 89.7% and 80.6%, respectively. The research provides theoretical basis for the popularization of Rubus suavissimus S. industry and the development of new natural antioxidants in Guangxi.
