摘要
目的对从软珊瑚Sarcophytonsp.中发现的青霉属真菌Penicillium citrinum(15XS24ZM-2)的次级代谢产物进行研究。方法利用硅胶柱色谱、薄层色谱、ODS中压色谱、Sephadex LH-20凝胶柱色谱、高效液相色谱(HPLC)等方法对该菌的次级代谢产物进行了分离纯化;通过核磁共振(NMR)、质谱(MS)等方法,并结合文献对照,鉴定化合物的结构。结果从青霉属真菌Penicillium citrinum(15XS24ZM-2)中共发现8个化合物:2,3-dihydrosorbicillin(1)、citrinin(2)、penicitol(3)、dimethyl-4,7-dihydroxy-9-oxo-9H-xanthene-3,5-dicarboxylate(4)、emodin(5)、methyl-2,8-dihydroxy-6-methyl-9-oxo-9H-xanthene-1-dicarboxy(6)、8-hydroxy-6-methyl-9-oxo-9H-xanthene-1-carboxylate(7)、ergostane(8)。生物活性结果显示化合物3对HCT-116和HeLa细胞株有细胞毒性,IC50分别为25.5和12.3μmol/L。
Objective To investigate the secondary metabolites and their bioactivities of the Penicillium citrinum(15XS24ZM-2)isolated from a soft coral Sarcophyton sp..Methods The techniques of silica gel column,TLC,ODS medium-pressure chromatography,Sephadex LH-20 column chromatography and HPLC were used to isolate and purify the compounds from the secondary metabolites yield by Penicillium citrinum(15XS24ZM-2).The structures of the compounds were identified by NMR,MS and comparison with the data of literature.Results Eight compounds were isolated from the Penicillium citrinum(15XS24ZM-2)and identified as 2,3-dihydrosorbicillin(1),citrinin(2),penicitol(3),dimethyl-4,7-dihydroxy-9-oxo-9 H-xanthene-3,5-dicarboxylate(4),emodin(5),methyl-2,8-dihydroxy-6-methyl-9-oxo-9 H-xanthene-1-carboxylate(6),8-hydroxy-6-methyl-9-oxo-9 H-xanthene-1-carboxylate(7),ergostane(8).Bioactivity showed that compound 3 was cytotoxic to HCT-116 and HeLa cell lines and IC50 were 25.5,12.3μmol/L,respectively.
