大蒜素对脓毒症小鼠急性肝损伤的作用 被引量:7

Effect of allicin on sepsis-induced acute liver injury in mice

摘要 目的 探讨大蒜素对脓毒症小鼠急性肝损伤的作用及机制。方法 按照随机数字表将8周龄雄性Balb/c小鼠随机分为3组:假手术组、模型组和实验组,每组8只。用盲肠结扎穿刺法建立脓毒症小鼠模型。实验组在造模后用大蒜素(30 mg·kg-1,ip)干预,假手术组和模型组给予等量0. 9%Na Cl。以全自动生化分析仪检测血清谷丙转氨酶(GPT)和谷草转氨酶(GOT)活性;酶联免疫吸附法检测肝组织肿瘤坏死因子α(TNF-α)和白细胞介素-6(IL-6)水平;以免疫印迹检测肝组织中自噬蛋白微管相关蛋白1轻链3B(LC3B)和Beclin-1的表达。结果 正常组、模型组和实验组的血清GPT分别为(25. 65±4. 26),(195. 54±22. 65)和(95. 28±11. 74) U·L-1;这3组的GOT分别为(7. 42±1. 15),(86. 37±8. 53)和(40. 95±5. 28) U·L-1;这3组肝组织的TNF-α水平分别为(48. 26±10. 35),(213. 86±34. 38)和(103. 27±20. 28)pg·mg-1;这3组的IL-6水平分别为(72. 58±15. 65),(764. 38±75. 37)和(435. 36±54. 27) pg·mg-1;这3组的LC3 II/I相对表达量分别为0. 57±0. 12,1. 59±0. 18和3. 85±0. 45;这3组的Beclin-1的相对表达量分别为0. 46±0. 08,1. 08±0. 15和2. 36±0. 25。以上指标:模型组与正常组比较,差异均有统计学意义(P <0. 05,P <0. 01);实验组与模型组比较,差异均有统计学意义(P <0. 05,P <0. 01)。结论 大蒜素对CLP诱导的肝损伤具有一定的保护作用,其机制可能与增加自噬有关。 Objective To explore the effect and mechanism of allicin on acute liver injury in septic mice.Methods The male Balb/c mice aged8 weeks were randomly divided into three groups according by random number table:sham operation group,model group and experimental group.Sepsis was conducted by cecal ligation and puncture(CLP)and allicin(30 mg·kg-1,ip)was given post-CLP in experimental group.The sham and model group were given the same amount of saline.The levels of glutamic-pyruvic transaminase(GPT)and glutamic-oxaloacetic transaminase(GOT)in serum were detected by automatic analyzer.The levels of tumor necrosis factor alpha(TNF-α)and interleukin-6(IL-6)in liver tissues were detected by enzyme-linked immunosorbent assay.The expression of microtubule-associated protein 1 light chain 3 B(LC3 B)and Beclin-1 in liver tissue was detected by Western blotting.Results The serum levels of GPT in sham operation group,model group,and experimental group were(25.65±4.26),(195.54±22.65)and(95.28±11.74)U·L-1,respectively;the GOTlevels in the 3 group were(7.42±1.15),(86.37±8.53)and(40.95±5.28)U·L-1,respectively;the liver TNF-αlevels in the 3 groups were(48.26±10.35),(213.86±34.38)and(103.27±20.28)pg·mg-1,respectively;the IL-6 levels in the 3 groups were(72.58±15.65),(764.38±75.37)and(435.36±54.27)pg·mg-1,respectively;the relative expression of LC3 II/I in the 3 groups was 0.57±0.12,1.59±0.18,and 3.85±0.45,respectively;the relative expression of Beclin-1 in the 3 groups was 0.46±0.08,1.08±0.15 and 2.36±0.25,respectively.Significant difference was found between sham group and model group(P<0.05,P<0.01),and significant difference was also found between experimental group and model group(P<0.05,P<0.01).Conclusion Allicin may ameliorate sepsis-induced acute liver injury in mice by enhancing the level of autophagy.
