灯盏细辛抑制视网膜肿瘤坏死因子-α表达保护青光眼大鼠视觉功能 被引量:6

Erigeron breviscapus protects visual function in glaucoma rats by inhibiting retinal tumor necrosis factor-α expression

摘要 目的 研究灯盏细辛对青光眼大鼠视觉功能的影响及相关的作用机制。方法 将24只SD大鼠随机分为假手术组、模型组和实验组,每组8只。用Akira法构建青光眼模型,假手术组未烙闭大鼠双眼巩膜静脉;建模成功后,实验组肌肉注射1. 5 mg·kg-1灯盏细辛注射液,假手术组和模型组均肌肉注射等量0. 9%氯化钠,每天1次,连续干预30 d。用眼压计测量青光眼大鼠建模后眼压变化,用荧光金逆行标记法观察视网膜神经节细胞(RGCs)密度,用Western blot检测大鼠视网膜肿瘤坏死因子-α(TNF-α)表达情况。结果 假手术组、模型组和实验组给药后第4周眼压分别为(7. 96±0. 82),(21. 61±1. 34),(13. 25±0. 56) mm Hg;大鼠视网膜RGCs分别为(193. 21±30. 25),(93. 43±22. 31),(115. 21±33. 27)个/视野;视网膜TNF-α蛋白相对表达量分别为0. 87±0. 14,1. 49±0. 12,1. 02±0. 09。模型组的上述指标分别与假手术组、实验组比较,差异均有统计学意义(均P <0. 05)。结论 灯盏细辛可降低青光眼大鼠高眼压及视网膜TNF-α蛋白表达水平,对RGCs具有较好的保护作用。 Objective To study the effect of erigeron breviscapus onvisual function in glaucoma rats and its related mechanisms.Methods A total of 24 SD rats were randomly assigned to sham-operation group,model group and test group according to their body weight,with 8 rats in each group.The glaucoma model was established by Akira method,and the bilateral scleral veins were not branded in the sham-operation group.After the successful establishment of the model,the test group was intramuscularly injected with 1.5 mg·kg-1 erigeron breviscapus injection,and the sham-operation group and the model group were intramuscularly injected with the same amount of 0.9%NaCl.All groups received treatment once daily for 30 d.Intraocular pressure was measured by tonometer after the establishment of glaucoma rat model.The retinal ganglial cells(RGCs)density was observed by fluorescence gold retrograde labeling and fluorescence microscope.Tumor necrosis factor-α(TNF-α)expression was detected by immunofluorescence and Western blot(WB).Results The intraocular pressure(IOP)in sham-operation group,model group and test group were(7.96±0.82),(21.61±1.34),(13.25±0.56)mmHg at the 4th week after administration;the RGCs in rat retina were(193.21±30.25),(93.43±22.31),(115.21±33.27)per field of vision;and the relative expression of TNF-αprotein in retina were 0.87±0.14,1.49±0.27 and 1.02±0.09,respectively.The above indexes of model group were significantly different from those of sham operation group and experimental group(all P<0.05).Conclusion Erigeron breviscapus can reduce the high intraocular pressure and the expression of retinal TNF-αprotein in glaucoma rat and has a better protective effect on RGCs.
