北五味子中总木脂素类成分的纯化工艺及抗氧化作用研究 被引量:11

摘要 目的:以五味子醇甲、醇乙、酯甲、甲素、乙素为指标成分建立高效液相色谱分析方法,通过考察乙醇浓度、料液比、提取次数、提取时间等因素优化提取工艺,并筛选出最佳提取纯化工艺为。方法:用95%乙醇料液比为1∶8回流提取1 h,连续提取两次得五味子总木脂素粗提液;再用HPD100大孔树脂进行纯化,0.15 g/mL的药液浓度进行上样,上样量最大为7 BV,径高比1∶6,11 BV 95%乙醇洗脱,上样流速和洗脱流速均为2 BV/h,得到提取纯化后总木脂素的含量占冻干后固形物的60%以上,并且该工艺具有效率高、成本低、纯度高等优点。结果:抗氧化活性研究方面,与同等浓度的VC对照品相比,北五味子总木脂素类成分的总抗氧化能力是Vc的1/7左右,清除超氧阴离子自由基的强度是VC的1/6左右。结论:说明其具有良好的抗氧化活性,为进一步研究及开发北五味子提供理论支持。 Objective:Five representative components of lignin(Schisandra alcohol A,Schisandra alcohol B,Schisandra Ester A,Schisandra methyl,Schisandra ethyl)were used as the indexes.The high performance liquid phase was used as the analysis method.The best condition for extraction of total lignin in Schisandra was to observe the effect of ethanol concentration,extraction times and extraction time.Methods:The optimized extraction and purification process was as follows:1 h was extracted with the ratio of 1∶8 for 95%alcohol mixture.The total lignin extract was extracted twice,and then purified with HPD100 macroporous resin.The concentration of the samlpe was 0.15 g/mL.The maximum sample quantity was 7 BV and the diameter ratio was 1∶6 and 11 BV 95%ethanol for elution.Both the upper flow rate and the elution flow rate were 2 BV/h.The content of total lipids after extraction and purification could be obtained after freezing.The total amount of solids can reach more than 60%.The optimized extraction and purification process of total lignin in Schizophyllum had the advantages of high efficiency,low cost and high purity.Results:In terms of antioxidant activity,the total antioxidant capacity of total lignans of Schisandra was about 1/7 of the same concentration of Vc reference.The intensity of scavenging superoxide anion radicals was 1/6 of VC reference.Conclusions:So it had good antioxidant activity.And it will provide theoretical support for further research and development of Schisandra.
