摘要
目的观察芪地糖肾颗粒对糖尿病肾病(diabetic nephropathy, DN)大鼠肾组织内转换生长因子(TGF)-β1-Smad2/3信号通路的影响。方法采用单侧腹腔注射链脲佐菌素(streptozotocin, STZ)制备DN大鼠模型,并用中药芪地糖肾颗粒干预。ELISA法检测24 h尿微量白蛋白(microalbuminuria, MAU)水平,光学显微镜观察大鼠肾脏结构,Western Blot检测大鼠肾组织TGF-β1、smad2、smad3、a-SMA蛋白表达。结果中药芪地糖肾颗粒干预后,大鼠24 hMAU水平下降(P<0.01);肾组织内 TGF-β1表达水平降低(P<0.01);光镜下模型组大鼠肾小球系膜基质增多、毛细血管基底膜增厚,肾小管上皮细胞肥大扩张,呈空格样改变,中药干预后肾小管病变程度降低。结论芪地糖肾颗粒通过TGF-β1-smad2/3通路下调TGF-β1的表达水平来防治糖尿病肾病的纤维化。
Objective?To observe the effect of Qidi tangshen Granule on the transforming growth factor (TGF)-β1-Smad2/3 signaling pathway in renal tissue of diabetic nephropathy (DN) rats. Methods The DN rat model was prepared by unilateral intraperitoneal injection of STZ and intervened with Qidi Tangshen Granule. The 24h urinary microalbumin level was detected by ELISA. The kidney structure was observed by light microscopy. The expression of TGF-β1, Smad2, Smad3, and a-SMA protein in kidney tissue was detected by Western Blot. Results After intervention with Qidi Tangshen Granule, the urinary microalbumin level decreased in 24 hours (P<0.01);the expression of TGF-β1 in kidney tissue decreased (P<0.01);the glomerular mesangial matrix in rat model group under light microscope increased, capillary basement membrane thickened, renal tubular epithelial cells showed hypertrophy and expansion with a space-like change, the degree of renal tubular disease after intervention of Qidi tangshen Granule reduced. Conclusion Qidi tangshen Granule can prevent fibrosis of diabetic nephropathy by down-regulating the expression of TGF-β1 through TGF-β1-smad2/3 pathway.
