Expression and Clinical Significance of Talin and Vinculin Proteins in Colon Cancer Tissues

摘要 目的分析踝蛋白(Talin)和纽蛋白(Vinculin)在结肠癌(colorectal cancer,CRC)组织中的表达及意义。方法收集80例经病理学证实为CRC患者的组织标本及其癌旁(距癌组织≥5 cm)正常组织标本。比较CRC组织和癌旁正常组织Talin、Vinculin蛋白表达情况,并分析与CRC患者临床病理特征及生存情况的关系。结果 Talin和Vinculin在CRC组织中表达率分别为27. 50%,21. 25%,表达率均明显低于癌旁正常组织(77. 50%、85. 00%),差异有统计学意义(P <0. 05);Talin、Vinculin在不同性别、年龄及肿瘤直径中的表达无统计学意义(P均> 0. 05),淋巴结未转移及TNM分期Ⅰ+Ⅱ期病例Talin、Vinculin表达阳性率明显高于淋巴结转移及TNM分期Ⅲ+Ⅳ期病例,差异有统计学意义(P <0. 05),高分化和中分化腺癌Talin、Vinculin表达阳性率明显高于低分化腺癌(P <0. 05);Spearman相关分析显示,在CRC组织中,Talin和Vinculin表达之间呈高度正相关(γ=0. 52,P <0. 01);80例患者随访3年死亡22例,3年生存率为72. 50%(22/80),CRC组织中Talin及Vinculin阳性表达患者3年生存率分别90. 91%和94. 12%,明显高于低表达患者(65. 52%,66. 67%),差异有统计学意义(P <0. 05)。结论 Talin、Vinculin在CRC组织中均呈低表达,二者可作为CRC患者预后的保护因素,此外,Talin、Vinculin与CRC生物学行为有关,可能成为CRC诊断的潜在指标。 Objective To analyze the expression and significance of Talin and Vinculin in colorectal cancer( CRC) patients. Methods Tissue specimens of 80 patients with pathologically confirmed CRC and normal tissues of adjacent tissues( ≥5 cm from cancer tissue) were collected. Talin and Vinculin protein expression in CRC tissues and paracancer normal tissues were compared,and the relationship between Talin and Vinculin protein expression and clinicopathological characteristics and survival of CRC patients was analyzed. Results The expression rates of Talin and Vinculin in colon cancer tissues were 27. 50% and21. 25%,respectively,which were significantly lower than those of normal paracancer tissues( 77. 50% and 85. 00% %),and the differences were statistically significant( P < 0. 05). Talin and Vinculin expression in different sex,age and tumor diameter had no statistical significance( P > 0. 05),Talin and Vinculin expression in lymph node transfer and TNM stageⅠ + Ⅱ cases were significantly higher than the lymph node metastasis,and TNM stage Ⅲ + Ⅳ cases,the difference was statistically significant( P <0. 05). Talin and Vinculin expression were significantly higher in highly differentiated and moderately differentiated adenocarcinoma patients than of low-differentiated adenocarcinoma patients( P < 0. 05). Spearman correlation analysis showed that there was a highly positive correlation between Talin and Vinculin expression in colon cancer tissues( γ = 0. 52,P < 0. 01). 22 patients died among the 80 patients during 3 years of follow-up and the 3-year survival rate was 72. 50%( 22/80). 3-year survival rates of Talin and Vinculin positive expression patients in colon cancer tissues were 90. 91% and 94. 12%,respectively,which was significantly higher than those of low expression patients( 65. 52%,66. 67%),and the difference was statistically significant( P < 0. 05).Conclusion Talin and Vinculin are both low expressed in colon cancer tissues,which can be used as protective factors for the prognosis of patients with colon cancer. In addition,Talin and Vinculin are related to the biological behavior of colon cancer,and may be potential indicators for the diagnosis of colon cancer.
