健脾补肾解毒方对Lewis肺癌小鼠脾脏CD+4CD+25调节T细胞含量的影响

Effect of Jianpi Bushenjiedu Recipe on the expression of CD+4CD+25T cells in mice spleen with Lewis lung cancer

目的使用动物实验研究探索健脾补肾解毒方通过下调lewis肺癌小鼠脾脏中的CD4^+CD25^+调节性T细胞的比例来达到抑制肿瘤的作用机制。方法将40只C57BL/6J小鼠,随机分为正常组、荷瘤模型组、化疗组(顺铂组)、中药组(健脾补肾解毒方)、联合组(健脾补肾解毒方+顺铂组),给药14 d后处死小鼠,剥离脾脏,用流式细胞仪检测小鼠脾脏CD4^+CD25^+的含量。结果小鼠脾脏CD4^+CD25^+调节T细胞含量与正常组比较,中药组、荷瘤模型组、化疗组比率明显升高,差异有统计学意义(P<0.05);联合组比率虽高于正常组,但差异无统计学意义(P>0.05);与荷瘤模型组比较,化疗组、中药组、联合组比率明显降低,差异有统计学意义(P<0.05)。结论健脾补肾解毒方通过下调lewis肺癌小鼠脾脏中CD4^+CD25^+调节性T细胞的比例能达到抑制肿瘤的作用。

Objective To explore the mechanism of Jianpi Bushen Jiedu recipe in inhibiting tumors by reducing the proportion of CD4^+CD25^+ regulatory T cells(Treg) in the spleen of Lewis lung cancer mice through animal experiments.Methods A total of 40 C57 BL/6 J mice were randomly divided into normal group,model group,chemotherapy group(cisplatin group),traditional Chinese medicine group(Jianpi Bushen Jiedu recipe),combined group(Jianpi Bushen Jiedu recipe + cisplatin group). After 14 days,the mice were executed and their spleens were stripped. The content of CD4^+CD25^+ in mouse spleens were detected by flow cytometry.Results Compared with the normal group,the ratio of CD4^+CD25^+ regulation T cells in traditional Chinese medicine group,model group,chemotherapy group was significantly higher(P<0.05),although the ratio in combined group was higher than that in the normal group,but the difference was not statistically significant(P>0.05). Compared with the model group,the chemotherapy group,the tradition Chinese medicine group,the combined group was significantly lower,the difference was statistically significant(P<0.05).Conclusion Jianpi Bushen Jiedu recipe can inhibit the tumor by decreasing the proportion of CD4^+CD25^+regulatory T cells in the spleen of Lewis lung cancer mice.
