紫外诱导对火木层孔菌次级代谢的影响

摘要 目的通过诱导改变火木层孔菌的次级代谢产物,考察次级代谢产物中是否有桑黄素类成分产生。方法通过紫外照射火木层孔菌,挑取致死率在95%以上的优势菌株进行液体发酵培养,以桑黄素Ⅰ和二聚桑黄素A作为对照,通过HPLC-DAD和HPLC-MS/MS检测代谢产物。结果发现利用紫外照射火木层孔菌进行诱导23 min时,达到火木层孔菌95%的致死率,从中挑取优势诱导菌株8株,对这8株菌株的液体发酵次级代谢进行分析,发现UV23-7能够产生含量和数量有所增加的桑黄素类成分。结论利用紫外诱导方法改变火木层孔菌的次级代谢这一方法可行。 Objective To investigate whether secondary metabolites of P. igniarius were induced and to detect the presence of Phellinus igniarius compounds in secondary metabolites.Methods The dominant strain irradiated by UV with a mortality rate above 95% was selected for liquid fermentation and the strain’s metabolites were detected by HPLC-DAD and HPLC-MS/MS,the phelligridinⅠand phelligridimer A were used as the control group.Results It was found that the 95% lethality of P. igniarius was induced by ultraviolet irradiation for 23 minutes,and 8 strains of dominant strains were picked from them. The liquid fermentation of 8 strains were analyzed,the results showed the induced strain UV23-7 produced the increased content and quantity of Sang flavins.Conclusion It is feasible to change the secondary metabolism of P. igniarius. by UV induction.The method of ultraviolet induction could change the metabolism of P. igniarius.
