枸杞多糖对弱精精子冻后洗涤过程中的保护作用 被引量:2

摘要 目的探讨在弱精精子冻后的优选过程中枸杞多糖对精子功能的影响。方法选取22例男性弱精患者,精液样本冷冻复苏后分为2组:SW组(冻后洗涤组)和SW+LBP组(冻后枸杞多糖干预洗涤组)。精液经常规冷冻后,采用非连续密度梯度法处理冻后精子,其中SW+LBP组的离心液中加入一定浓度的枸杞多糖,最终检测精子的前向运动率(PR)、存活率、正常形态率、精子DNA断裂指数(DFI)以及离心过程中2组间丙二醛(MDA)、活性氧(ROS)的含量,并进行比较分析。结果 PR检测结果显示,SW+LBP组高于SW组;DFI检测结果显示,SW+LBP组显著低于SW组。MDA、ROS检测结果显示,SW+LBP组显著低于SW组。精子存活率和正常形态率结果显示,2组之间差异无统计学意义。结论枸杞多糖可以减少精子冻后优选过程中精子的氧化应激,提高弱精精子优选后的精子活力及精子功能。 Objective To explore the effect of Lycium barbarum polysaccharide on sperm function in the optimization of asthenospermia sperm after freezing.Methods 22 male patients with asthenospermia were selected according to the standard of the fifth edition of the World Health Organization Laboratory Manual for Human Semen Examination and Treatment.After cryopreservation and resuscitation,the semen samples were divided into two groups:SW group(post-freeze washing group)and SW+LBP group(post-freeze Lycium barbarbarum polysaccharide intervention washing group).After the semen was frozen regularly,the frozen sperm was treated by discontinuous density gradient method.A certain concentration of Lycium barbarum polysaccharide was added into the centrifuge of the intervention group.The sperm forward motility(PR),survival rate,normal morphology rate,sperm DNA fragmentation index(DFI),malondialdehyde(MDA)and ROS between the two groups during centrifugation were detected.The results were compared and analyzed.Results PR test results showed that SW + LBP group was significantly higher than SW group,DFI test results showed that SW+ LBP group was significantly lower than SW group).The results of MDA and ROS showed that SW+LBP group was significantly lower than SW group.The results of sperm viability and normal morphology showed that there was no significant difference between the two groups.Conclusion Lycium barbarum polysaccharide can reduce the oxidative stress of sperm in the process of sperm optimization after freezing,thus improving the motility and sperm function of asthenospermia sperm.
