叔丁基对苯二酚对大鼠视网膜新生血管的作用及其机制 被引量:3

摘要 目的探讨叔丁基对苯二酚对大鼠视网膜新生血管的作用及其机制。方法雄性Sprague Dawley大鼠30只随机分为3组(对照组、模型组与治疗组),每组10只。对照组为正常SD大鼠,给予基础饲料喂养;模型组与治疗组为氧诱导视网膜病变模型,治疗组在模型构建后7 d采取玻璃体注射叔丁基对苯二酚治疗。记录大鼠治疗前和治疗后第7天、第14天与第21天体重;观察大鼠视网膜无灌注区铺片面积和新生血管内皮细胞数量和视网膜病理情况;检测大鼠核因子E2相关因子2(Nrf2)、血管内皮生长因子(VEGF)蛋白相对表达量。结果模型组和治疗组造模成功后(治疗前)和治疗后第7天、第14天与第21天的体重均显著低于对照组(P<0.05),治疗前和治疗后第7天模型组和治疗组大鼠体重比较差异无统计学意义(P>0.05),治疗后第14天与第21天治疗组大鼠的体重显著高于模型组(P<0.05)。对照组大鼠视网膜未见无灌注区及新生血管内皮细胞出现,治疗组大鼠视网膜无灌注区铺片面积和新生血管内皮细胞数量均显著少于模型组大鼠(P<0.05)。与模型组相比,治疗组的视网膜层次病理性变化减弱,水肿程度降低,各层的结构稀疏排列的程度降低。模型组与治疗组大鼠的Nrf2、VEGF蛋白相对表达量均显著高于对照组(P<0.05),治疗组Nrf2、VEGF蛋白相对表达量均显著低于模型组(P<0.05)。结论叔丁基对苯二酚治疗的可促进视网膜病变模型大鼠体重恢复,并抑制新生血管内皮细胞的生成和Nrf2、VEGF蛋白的表达,减轻视网膜损伤。 Objective To investigate the effects of tert-butyl hydroquinone on retinal neovascularization and its mechanism.Methods 30 male Sprague Dawley rats were randomly divided into three groups(control group,model group and treatment group),with 10 rats in each group.The control group was normal SD rats,and were fed with the basal diet.The model group and the treatment group were oxygen-induced retinopathy models.The treatment group was treated with vitreous injection of tert-butyl hydroquinone 7 days after the model was constructed.Rat body weight was recorded before treatment and on days 7 th,14 th and 21 st after treatment.The area of the non-perfused area of the rat retina and the number of neovascular endothelial cells and pathological changes of the retina were observed.The relative expression levels of Nrf2 and VEGF proteins in rats were detected.Results Compared with the control group,the body weights of the model group and the treatment group before treatment and on the 7 th,14 th and 21 st day after treatment were significantly lower(P<0.05).There was no significant difference in body weight between the model group and the treatment group before treatment and on the 7 th day after treatment(P>0.05).The body weight of the rats in the treatment group on the 14 th day and the 21 st day after treatment was significantly higher than that in the model group(P<0.05).There was no perfusion area and neovascular endothelial cells in the retina of normal control group.The area of retinal non-perfusion area and the number of neovascular endothelial cells in the treatment group were significantly lower than those in the model group(P<0.05).Compared with the model group,the pathological changes of the retina level in the treatment group were weakened,the degree of edema was reduced,and the degree of sparse arrangement of the layers was reduced.The relative expressions of Nrf2 and VEGF protein in the model group and the treatment group were significantly higher than those in the control group(P<0.05).The relative expressions of the above two indicators in the treatment group were significantly lower than those in the model group(P<0.05).Conclusion Tert-butyl hydroquinone treatment can promote the recovery of body weight in rats with retinopathy,and inhibit the formation of vascular endothelial cells and the expression of Nrf2,VEGF protein,and also can reduce retinal damage.
