摘要
【目的】研究Kruppel样转录因子5(Kruppel like factors 5,KLF5)在胰腺癌组织中的表达情况和它的表达与疾病临床病理特征的关系,评价其对胰腺癌患者预后的影响,并分析KLF5在胰腺癌发生、发展中可能的病理机制。【方法】收集整理GEO与TCGA数据库资料;分析KLF5在胰腺癌与正常胰腺组织表达情况的差异,利用t检验统计KLF5的表达与胰腺癌临床病理特征之间的关系;采用基因富集分析法(gene set enrichment analysis,GSEA)预测KLF5相关的基因通路。【结果】在GSE28735数据集中,KLF5在胰腺癌中的表达明显高于正常的胰腺组织,KLF5的表达与肿瘤的分级、TNM分期存在相关性(P<0.05);在TCGA数据集中,高表达KLF5的胰腺癌患者总体生存率明显低于低表达组(P=0.002);GESA结果显示,KLF5可能富集到E2F通路、G2/M检查点、MYC-V1通路、MYC-V2通路、MTORC1通路、有丝分裂通路、DNA修复通路、糖酵解通路相关的基因集。【结论】胰腺癌组织中KLF5的表达高于正常胰腺组织,KLF5的表达与胰腺癌的分级、TNM分期有关,高表达KLF5的胰腺癌患者临床预后不良,有可能作为胰腺癌患者肿瘤进展的标志物和治疗胰腺癌的新的分子靶标。
【Objective】To explore the expression of Kruppel like factor 5(KLF5)in pancreatic cancer tissues and its relationship with the clinicopathological characteristics of the disease,evaluate its effect on the prognosis of pancreatic cancer and analyze the possible pathological mechanism of KLF5 in the occurrence and development of pancreatic cancer.【Methods】GEO and TCGA databases were collected and sorted out.The difference of KLF5 expression in pancreatic cancer and normal pancreatic tissues were analyzed,and the relationship between KLF5 expression and clinicopathological characteristics of pancreatic cancer was statistically analyzed by t test.Gene enrichment analysis(GSEA)was used to predict KLF5 related gene pathways.【Results】The expression of KLF5 in pancreatic cancer was significantly higher than that in normal pancreatic tissues,and the expression of KLF5 was statistically significant with tumor grade and TNM stage(P<0.05)in the GSE28735 data set.Patients with high expression of KLF5 had significantly shorter overall survival than those with low expression(P=0.002)in the TCGA data set.The results of GESA showed that KLF5 might regulate gene sets related to E2 F pathway,G2/M checkpoint,MYC-V1 pathway,MYC-V2 pathway,MTORC1 pathway,mitotic pathway,DNA repair pathway and glycolysis pathway.【Conclusion】The expression of KLF5 in pancreatic cancer tissues is higher than that in normal pancreatic tissues,and the expression of KLF5 is related to the grade and TNM stage of pancreatic cancer.Patients with pancreatic cancer and high expression of KLF5 have poor clinical prognosis,which may be used as a potential biomarker for the tumor progression in pancreatic cancer patients and a novel molecular target for the treatment of pancreatic cancer.
