基于GEO数据库对脑梗死相关差异基因的分析研究 被引量:1

摘要 目的比较模型组(MCAO组)大鼠与假手术组(Sham组)大鼠脑组织的差异基因,分析差异基因影响的功能通路,为脑梗死的治疗提供可行的研究方向和具体的作用靶点。方法在GEO数据库下载MCAO组大鼠与Sham组大鼠脑组织中的表达谱芯片,并通过R软件得到差异基因;利用DAVID6.8分析差异基因的GO功能富集和KEGG通路富集。结果通过分析得到148个差异表达基因,123条GO富集通路,主要涉及药物反应、脂多糖反应、炎症反应、有机环状化合物反应及细胞外基质等生物过程,并与蛋白结合、受体结合、蛋白质同源化活性、整合素结合、蛋白酶结合及趋化因子的活性等分子功能相关;KEGG通路富集分析,得到显著性富集通路23条,涉及TNF信号通路、NF-kB信号通路、Toll样受体信号通路和HIF-1信号通路等。结论TNF信号通路与NF-kB信号通路可能是治疗脑梗死的行之有效的干预方向。 Objective To compare the gene expression differences in MCAO rats and Sham rats,and to analyze the functional pathways.The purpose was to provides a feasible research direction and specific target for the treatment of cerebral infarction.Methods We downloaded the data about the brain tissues of MCAO rats and Sham rats from the GEO database and obtained ifferentially expressed genes through R software.Finally,we obtained GO enrichment analysis and KEGG pathway analysis by using DAVID 6.8.Results We obtained 148 differentially expressed genes and 123 GO enrichment pathways,including the main biological processes were drug reaction,lipopolysaccharide reaction,inflammatory reaction,organic cyclic,compound reaction and extracellular matrix and so on.At the same time,It is also associated with protein binding receptor binding protein homologous activity integrin-binding protease binding and chemokine activity.The enrichment analysis of KEGG pathway showed that there were 23 significantly enriched pathways,involving TNF signaling pathway,NF-kB signaling pathway,Toll-like receptor signaling pathway and HIF-1 signaling pathway.Conclusion It may be a effective intervention directions for TNF signaling pathway and NF^kB signaling pathway to treat the cerebral infarction.
