摘要
目的基于网络药理学探讨黄英-山药药对治疗2型糖尿病的作用机制。方法通过中药系统药理学数据库和分析平台(TCMSP)检索黄茂山药药对中药物成分,并以口服生物利用度(OB)值$30%、类药性(DL)值M 0.18为条件筛选出黄茂山药的有效活性成分,确定药物有效活性成分后通过TCMSP数据库预测相应靶点,再将预测的靶点输入到Uniprot中转换为相应的基因。将有效活性成分及靶点对应的基因导入Cytoscape3.2.1中构建黄茂-山药有效成分一靶点网络。通过PharmGkb数据库检索2型糖尿病相关基因,并将检索出的基因导入Cytoscape3.2.1,利用Bisogenet插件构建2型糖尿病疾病靶点网络。利用Cytoscape3.2.1软件中Meige插件将黄英-山药有效活性成分一靶点网络和疾病靶点网络进行合并。根据degree值筛选核心基因团,最后利用DAVID对筛选出的核心基因团进行GO注释和KEGG通路分析。结果黄茂有效活性成分20个,山药有效活性成分16个,2型糖尿病相关疾病基因43个.KEGG通路为PI3K/Akt信号通路、FoxO信号通路相关、甲状腺激素信号通路。结论黄蒂-山药的有效活性成分治疗2型糖尿病可能与PI3K/Akt信号通路、FoxO信号通路相关、甲状腺激素信号通路相关。
Objective To explore the mechanism of Astragalus-Chinese yam in the treatment of diabetes mellitus type 2(T2DM)based on network pharmacology.Methods Firstly,retrieving the herbal ingredients of Astragalus-Chinese yam through the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP),while screening the effective active ingredients of Astragalus-Chinese yam on the condition that the oral bioavailability(OB)value$30%and the drug-like(DL)value A 0.18.After determining the effective active ingredient of the herbs,predict the conesponding target point through the TCMSP database,and then input them into the Uniprot to convert the corresponding gene.And then,Importing effective active ingredients and target corresponding genes into Cytoscape3.2.1 software to build an effective active component of Astragalus-Chinese yam-Target network.Secondly,retrieving the genes related to Diabetes mellitus type 2 by the PharmGkb database,and import them into Cytoscape3.2.1, then constructing the disease target network of Diabetes mellitus type 2 throughBisogenet plug-in. Thirdly, merging the effective active component of effective active component of Astragalus-Chinese yam-Target network and the disease target network of Diabetes mellitus type 2 by the Merge plug-in inCytoscape3.2.1 software. Screening the core gene clusters according to degree value, finally, using the DAVID toanalyze the GO annotation and KEGG pathway of the selected core gene clusters. Results There were 20 activecomponents of Astragalus, 16 active components of Chinese yam and 43 genes related to Diabetes mellitus type 2.KEGG pathway is PI3K/Akt signal pathway, FoxO signal pathway correlation and thyroid hormone signal pathway.Conclusion the effective components of Astragalus-Chinese yam pair in the treatment of Diabetes mellitus type 2 maybe related to PI3K/Akt signaling pathway, FoxO signaling pathway and thyroid hormone signaling pathway.
