摘要
目的探讨类风湿关节炎(RA)患者血清miRNAs(miR-146a-5p、miR-155-5p和miR-124a-3p)的表达水平及其临床意义。方法收集2016年10月至2018年10月在我院门诊就诊的39例RA患者临床资料、全血及血清样本。全血样本用于检测血沉(ESR),血清样本用于检测类风湿因子(RF)、C反应蛋白(CRP)和miRNAs核酸。采用SYBR Green实时荧光定量PCR检测RA患者血清miR-146a-5p,miR-155-5p和miR-124a-3p的表达水平。结果RA患者治疗前血清miR-146a-5p(1.742±1.058)表达水平显著高于HC组(1.045±0.772),且与DAS28评分(r=0.8365,P=0.0045)、ESR值(r=0.4372,P=0.0325)和RF含量(r=0.7336,P=0.0137)呈正相关;而miR-155-5p(U=42.00,P=0.0329)和miR-124a-3p(U=44.5,P=0.0445)表达水平显著低于HC组,且miR-155-5p表达水平与RF含量呈负相关(r=-0.4453,P=0.0316),miR-124a-3p表达水平与DAS28评分(r=-0.5387,P=0.0258)和RF含量(r=-0.4365,P=0.0463)呈负相关;RA患者治疗后血清miR-146a-5p表达水平较治疗前显著下降(U=60.00,P=0.0038),miR-155-5p(U=64.00,P=0.0059)和miR-124a-3p(U=85.00,P=0.0422)表达水平较治疗前显著升高。结论RA患者血清miR-146a-5p、miR-155-5p和miR-124a-3p的表达水平异常,对RA患者的诊断具有潜在的临床应用价值。
Objective To investigate the expression levels and clinical significance of serum miRNAs,including miR-146a-5p,miR-155-5p and miR-124a-3p,in patients with rheumatoid arthritis(RA).Methods The clinical data,whole blood and serum samples from 39 RA patients hospitalized in our hospital during October 2016 and October 2018 were collected.Whole blood specimens were used to determine erythrocyte sedimentation rate(ESR),and serum samples were used to detect rheumatoid factor(RF),C-reaction protein(CRP)and miRNAs.The expression levels of serum miR-146a-5p,miR-155-5p and miR-124a-3p in RA patients were detected by SYBR Green real-time fluorescence quantitative PCR.Results The expression levels of serum miR-146a-5p(1.742±1.058)in RA patients before treatment were significantly higher than that in healthy controls(HC,1.045±0.772),which were positively correlated with DAS28 scores(r=0.8365,P=0.0045),ESR(r=0.4372,P=0.0325)and RF levels(r=0.7336,P=0.0137).However,the expression levels of serum miR-155-5p(U=42.00,P=0.0329)and miR-124a-3p(U=44.5,P=0.0445)in RA patients were significantly lower than that in HC,and the expression levels of serum miR-155-5p were negatively correlated with RF levels(r=-0.4453,P=0.0316),and the expression levels of serum miR-124a-3p were negatively correlated with DAS28 scores(r=-0.5387,P=0.0258)and RF levels(r=-0.4365,P=0.0463).After treatment,the expression levels of serum miR-146a-5p(U=60.00,P=0.0038)in RA patients were significantly decreased,while the expression levels of serum miR-155-5p(U=64.00,P=0.0059)and miR-124a-3p(U=85.00,P=0.0422)were significantly increased.Conclusion The abnormal expression levels of serum miR-146a-5p,miR-155-5p and miR-124a-3p in RA patients have potential clinical significance for the diagnosis of RA.
