摘要
[目的]以大葱花序轴作为外植体材料,研究不同培养基组成对大葱花序轴愈伤组织诱导、芽分化、继代和生根培养的影响。[方法]以花蕾作为外植体,采用不同浓度的6-BA和NAA组合配成8种MS培养基,筛选出诱导、分化、继代和生根培养较适合的培养基。[结果]MS+BA7.0 mg/L+NAA 0.01 mg/L对愈伤诱导的效果最好;MS+6-BA 0.5 mg/L+NAA 0.01 mg/L为分化培养基的适宜配方;MS+6-BA 0.4 mg/L+NAA 0.2 mg/L为继代培养基的适宜配方;MS+IAA 0.05 mg/L生根效果很好。[结论]为大葱花序轴组织培养提供参考依据。
[Objection]The aim was to study on tissue culture of welsh onion from rachis.[Method]The flower bud was used as the explant with eight different concentration mediums of 6-BA and NAA.The adaptive medium was screened of induction,differentiation and rooting culture.[Result]The suitable culture medium for rachis regeneration was MS+BA 7.0 mg/L+NAA 0.01 mg/L.MS+BA 0.5 mg/L+NAA0.1 mg/L was suitable formulations for differentiation medium.MS+BA 0.4 mg/L+NAA 0.2 mg/L was suitable formula for subculture medium.MS+IAA 0.05 mg/L was suitable for rooting well.[Conclusion]The study could provide references for welsh onion bud tissue culture.
