摘要 目的:野菊花多糖的提取工艺优化及其抗氧化活性评价。方法:在单因素试验的基础上,以料液比、提取时间、提取温度为自变量,以野菊花多糖的提取率为指标,采用Box-Behnken响应面法优选野菊花多糖的提取条件;并通过对DPPH·和·OH的清除效果来评价其体外抗氧化活性。结果:野菊花多糖最佳提取工艺为料液比:1∶25 g·mL-1,提取时间:2.5 h,提取温度:92.3℃,在此条件下多糖提取率为3.25%;Sevage法脱蛋白脱除率为23.58%;过氧化氢法脱色率为61.18%;纯化后的野菊花多糖主要在0.8μm陶瓷膜渗透液中。野菊花多糖对DPPH·和·OH具有较好的清除能力。结论:该优化工艺实现了野菊花多糖的高效提取,微滤膜可以实现对野菊花多糖的分离纯化。 Objective:To explore the optimum extraction process of polysaccharides from Chrysanthemum indicum and evaluate its antioxidant activity.Methods:Based on single factor experiment,solid-liquid ratio,extraction time and extraction temperature were variables,and the extracting ratio of polysaccharides was used to evaluate the extraction process.Response surface methodology of Box-Behnken design was used to optimize the extraction conditions of total polysaccharides from Ch.indicum.Meanwhile,the antioxidant activity of polysaccharides was assessed by scavenging assays of DPPH·and·OH.Results:The optimal extraction process was as follows:liquid-solid ratio of 1∶25 g·mL-1,extraction temperature of 92.3℃,extraction time of 2.5 h,the extraction ratio of polysaccharides from Ch.indicum was 3.18%under the optimal conditions.The protein removal rate by Sevage method was 23.58%,the decolorization rate of H 2 O 2 was 61.18%,much of the polysaccharides were detected in the liquid penetrated from the ceramic membrane of 0.8μm.The polysaccharides from Ch.indicum could effectively scavenge DPPH·and·OH.Conclusion:The polysaccharides from Ch.indicum have been extracted efficiently by the above optimized technology.The preferred aperture of microfiltration can purify Ch.indicum polysaccharides.
