摘要
目的:响应面法优选山蜡梅叶颗粒中总黄酮的提取工艺。方法:以总黄酮得率为评价指标,在单因素试验的基础上,选取乙醇体积分数、液料比和提取时间为主要因素,通过Box-Behnken设计方案,建立总黄酮得率的二次回归方程,以Design-Expert 8.05软件对实验数据作统计处理,优选确定最佳提取条件。结果:山蜡梅叶颗粒中总黄酮最佳提取工艺条件为乙醇体积分数52%,液料比42∶1,超声时间60 min,总黄酮提取得率为0.447%。结论:优选得到的山蜡梅叶颗粒中总黄酮的提取工艺准确可行,质量稳定。
Objective:To Optimize the extraction technology of total flavonoids from Shanlameiye Granules.Methods:Response surface models were used to optimize the extraction technology of total flavonoids from Shanlameiye Granules.The total flavonoids were extracted using ultrasonic assisted ethanol extraction.On the basis of single factor,the concentration of ethanol,the amount of liquid added and the extraction time were the independent variables.Design-Expert response surface methodology was employed for selecting the optimum extraction process by the indexes of the contents of total flavonoids from Shanlameiye Granules.Results:The optimum extraction conditions for total flavonoids from Shanlameiye Granules were as follows,52.06%of the ethanol concentration,41.97 times of the amount of liquid added,extraction time 60.60 min.Conclusion:The extraction process obtained by response surface methodology was stable and reasonable,accurate and reliable.It was a feasible method to extract the total flavonoids from Shanlameiye Granules.
