摘要
目的探讨五味子7种木脂素单体对大鼠离体胸主动脉条张力的影响.方法制备离体血管条,采用去氧肾上腺素(phenylephrine,PE)预收缩血管条,运用张力检测法观察五味子木脂素单体对大鼠离体胸主动脉条张力的影响.结果五味子甲素、五味子乙素、五味子丙素、五味子酯甲、五味子醇甲、五味子醇乙及安五脂素对静息状态下胸主动脉条张力均无明显影响;用去氧肾上腺素(PE)预收缩胸主动脉条,发现五味子醇甲、五味子酯甲和五味子醇乙具有显著的舒张血管作用,对内皮完整型血管条的舒张率最大分别为(52.79±1.43)%、(43.56±2.17)%和(33.04±2.21)%(P<0.05或P<0.01或P<0.001);五味子甲素有微弱的内皮依赖性舒张血管作用(P<0.05或P<0.01);安五脂素舒张血管作用不显著,且非内皮依赖性较内皮依赖性明显;而五味子乙素和五味子丙素对血管条张力无明显影响.结论五味子木脂素发挥舒张血管作用的成分可能主要为五味子酯甲、五味子醇甲和五味子醇乙.
Objective To explore the effects of seven lignan monomers of Schisandra chinensis on the tension of isolated thoracic aorta strips of rat. Method The isolated vascular strips were prepared,and the vascular strips were pre-contracted with phenylephrine( PE). The effect of Schisandra chinensis lignan monomer on the tension of isolated thoracic aorta strips of rat was observed by tension test. Results Schisandrin A,Schisandrin B,Schisandrin C,Schisantherin A,Schizandrol A,Schizandrol B and Anwuligan had no significant effects on the resting tension of aortic strips. After pre-contracted aortic strips with phenylephrine( PE),Schizandrol A,Schisantherin A and Schizandrol B had significant vasodilating effect,the maximum relaxation rate of the endothelium dependence vascular strip is( 52. 79 ± 1. 43) %,( 43. 56 ±2. 17) %,( 33. 04 ±2. 21) %( P <0. 05 or P<0. 01 or P<0. 001);Schisandrin A has weak endothelium-dependent vasodilation( P <0. 05 or P <0. 01);The effect of Anwu ligan on vasodilation was not significant,and the endothelium independence was more obvious than endothelium dependence;While Schisandrin B and Schisandrin C had no obvious effect on the tension of aortic strips. Conclusion The components of Schisandra chinensis lignans exerting effect of vasodilation may be mainly Schizandrol A,Schisantherin A and Schizandrol B.
