摘要
目的探讨重组家蚕素基因(MBbxⅡ)稳定转染HepG2细胞后对细胞的生物学作用.方法将MBbxⅡ基因稳定转染于HepG2细胞中,以转染空质粒及未进行转染的HepG2细胞作为空白对照组,分别应用MTT方法检测细胞生长率,流式细胞术检测细胞周期.结果稳定转染MBbxⅡ基因的HepG2细胞增殖能力增强,且细胞多处于G2和S期.结论 MBbxⅡ促进了HepG2细胞增殖,可为研究MBbxⅡ在哺乳动物细胞中的胰岛素样作用奠定基础.
Objective The biological effect of stable transfection of recombinant Bombyxin II( MBbx II) gene on the proliferation of HepG2 cells were investigated,which laid a foundation for the effect of MBbx II on the insulinlike function of mammalian cells. Method HepG2 cells stably transfected with MBbx II gene were resuscitated.The transfected blank plasmid and non-transfected HepG2 cells were used as blank control group. The growth rate of cells was measured by MTT method. Cell cycle was detected by flow cytometry. Results The proliferation of HepG2 cells stably transfected with MBbx II gene was enhanced. Most of them were in G2 and S phases.Conclusion MBbx II could promote the proliferation of mammalian hepatocytes HepG2,which provides a basis for the islet like function of mammalian cells.
