陀螺果种子粕中皂苷提取及抗氧化活性 被引量:3

Extracting Saponin from Dross of Melliodendron Xylocarpum Seeds and Antioxidant Activity

摘要 以陀螺果种子榨油糟粕中的皂苷为研究对象,通过大孔树脂纯化吸附和酶法探索提高皂苷提取纯度方法.结果表明:添加纤维素酶有助于陀螺果种子糟粕中皂苷的溶出;当陀螺果种子糟粕物料粒度为80~100目,纤维素酶用量为1%,固液比为1∶25时,在常温下液体抽提1.5 h,皂苷溶出率可达4.19%;大孔吸附树脂AB-8对皂苷具有良好的吸附能力,最大吸附量为165 mg/g,用90%乙醇溶解可实现有效解吸.通过自由基清除法检测后发现,该皂苷具有抗氧化活性,且抗氧化能力随浓度增加而增加.研究结果可为皂苷提取分离工艺的建立提供技术依据,有助于提高陀螺果种子糟粕及精炼副产物的利用价值. The saponin in dross of Melliodendron Xylocarpum seeds was served as the research material. The extraction purity of M. Xylocarpum saponin was improved by enzyme method and macroporous resin to purify and adsorb at high quality. The results showed that: the addition of cellulase was conducive to the dissolution of saponin from the dross of M. Xylocarpum seeds. Under conditions of the grain size of the dross of M. Xylocarpum seeds as 80 ~ 100 orders,the dosage of cellulase at 1% level,the ratio of solid to liquid was 1 ∶ 25,and the liquid is extracted for 1. 5 h at room temperature,the dissolution rate of M. Xylocarpum saponin reached 4. 19%. The macroporous adsorption resin AB-8 had a good absorption capacity for saponin and the maximum adsorption capacity amounted to 165 mg/g,and the saponin can be dissolved in 90% ethanol to achieve effective desorption. By applying free radical scavenging method,the saponin showed possessing antioxidant activity and the antioxidant activity increased with the increase of the concentration. The results can provide the basis for the establishment of extraction and separation technology of saponin,and could help to improve the utilization value in dross of M. Xylocarpum seeds and its refined by-products.
