芽孢杆菌菌株TYF-B5-5的产乙醇特性及其全基因组测序分析 被引量:1

摘要 菌株TYF-B5-5(NCBI网站基因登录号为WJHD00000000)是太原理工大学微生物课题组从山西食醋中分离筛选出的1株嗜热耐酸野生菌株,通过NCBI数据库将菌株的16S rRNA进行Blast同源比对,确认为芽孢杆菌。菌株TYF-B5-5可以利用木质纤维素一步发酵生产乙醇,在以天然玉米秸秆为唯一碳源时,最适条件下乙醇浓度可达8.0 mmol/L。为了进一步了解该菌株的代谢机制及其特性,对菌株TYF-B5-5进行了全基因组测序。结果显示,该菌株基因组大小为3984415 bp,GC含量为43.56%,有11个scaffolds和12个contigs,共有4303个基因得到注释;通过数据库比对分析,完成了菌株TYF-B5-5丙酮酸到乙醇代谢过程5种关键基因的功能预测。菌株TYF-B5-5全基因组测序结果分析可为嗜热芽孢杆菌产乙醇代谢途径的研究提供理论支持。 The strain TYF-B5-5 is a strain of high-temperature resistant wild strain isolated from the Shanxi vinegar.The Blast homologous alignment of the 16S rRNA of the strain is confirmed by the NCBI database to be confirmed as Bacillus sp..The strain TYF-B5-5 can produce ethanol by one-step fermentation of lignocellulose.The yield of ethanol from straw as carbon source can reach 8.0 mmol/L at 45℃and pH 7.0.To further understand the metabolic mechanism and characteristics of the strain,the whole genome sequencing of the strain TYF-B5-5 was conducted.The results showed that the genome size was 3984415 bp,the GC content was 43.56%.There were 11 scaffolds and 12 contigs,and a total of 4303 genes were annotated,the functional prediction results of the key genes of the strain TYF-B5-5 pyruvate to ethanol was completed by database alignment analysis.The analysis results of the whole genome sequencing of strain TYF-B5-5 will provide theoretical support for the study of ethanol metabolism pathway of Bacillus thermophilus.
