白藜芦醇对脓毒症血管舒张功能的保护作用及机制 被引量:3

摘要 目的探讨白藜芦醇(Res)对脓毒症血管舒张反应性的保护作用及与eNOS和iNOS的关系。方法采用盲肠结扎穿孔复制脓毒症模型,将32只SD大鼠随机分为4组:正常对照组、脓毒症组、常规治疗组(乳酸林格液体复苏+血管活性药多巴胺+头孢呋辛钠)、Res治疗组(Res+常规治疗),每组8只。Res治疗组在模型后尾静脉给予白藜芦醇,12 h后行常规复苏并二次给药,观察4组大鼠肠系膜上动脉(SMA)的舒张反应性、肠系膜微血管动静脉流速、血流动力学、肝肾血流量、肠系膜上动脉eNOS和iNOS的表达及存活率和存活时间的变化。结果与正常对照组相比,脓毒症组大鼠血管舒张反应性明显降低;常规复苏可轻微改善脓毒症大鼠血管舒张反应性;Res可以明显改善脓毒症大鼠肠系膜血管舒张反应性,进一步研究发现,Res通过保护脓毒症大鼠的血管舒张反应性来改善血流动力学和组织器官的灌注量,提高了脓毒症大鼠的存活率和存活时间,机制研究发现Res对脓毒症大鼠血管舒张反应性的保护作用与血管eNOS和iNOS有关。结论白藜芦醇通过改善脓毒症大鼠的血管舒张反应性,从而实现了对整体器官的保护作用。 Objective To determine the protective effects of resveratrol(Res)on the vasodilatation function and its relationship with eNOS and iNOS.Methods The sepsis model of rats were established with cecal ligation and puncture(CLP).A total of 32 SD rats were randomly divided into normal control group,sepsis group,conventional treatment group(conventional fluid resuscitation+dopamine+cefuroxime,CT),Res+CT group(Res+conventional fluid resuscitation),8 rats in each group.Res+CT group were injected with Res by caudal vein.Conventional recovery was administered intravenously immediately and at 12 hours after CLP.The effects of resveratrol on vasodilatation function in superior mesenteric artery(SMA),the red blood cell velocity in mesenteric arteriole and venule,hemodynamics,liver and kidney blood flow,the expression of eNOS and iNOS and animal survive rates and survival time were observed.Results Compared with normal control group,vascular relaxation were significantly decreased after septic shock,which were slightly improved in conventional treatment group.Res could significantly improved the vasodilatation function in rats with sepsis,by which it could improve the hemodynamics and organ perfusion,the survive rates and survival time.The mechanism research showed that the protective effect of Res in vasodilatation of sepsis rats was related to the expressions of eNOS and iNOS.Conclusion Res can protect the hemodynamics and vital organ function by improving the vasodilatation reactivity.
