摘要
目的探讨比卡鲁胺对人三阴性乳腺癌MDA-MB-231细胞增殖的影响及其可能的机制。方法0、25、50、100μmol/L比卡鲁胺处理人三阴性乳腺癌MDA-MB-231细胞,MTT法检测细胞的抑制率;流式细胞仪检测比卡鲁胺对MDA-MB-231细胞作用后凋亡率及其细胞周期分布的影响。结果不同浓度(25、50、100μmol/L)比卡鲁胺处理后,MDA-MB-231细胞增殖能力明显下降,且存在时间、剂量依赖关系。比卡鲁胺可以明显地增加细胞凋亡,细胞周期表现为G0/G1期阻滞(P<0.05)。结论比卡鲁胺可能通过诱导细胞凋亡及抑制细胞周期的机制从而对人三阴性乳腺癌MDA-MB-231细胞的增殖产生抑制作用。
Objective To explore the effect of Bicalutamide on the proliferation of human triple negative breast cancer MDA-MB-231 cells and its potential molecular mechanism.Methods The human triple negative breast cancer MDA-MB-231 cells were treated with 0,25,50,100μmol/L Bicalutamide,MTT method was used to detect the inhibition rate of cells,and flow cytometry was performed to assay the effect of Bicalutamide on the apoptosis rate and cell cycle distribution of MDA-MB-231 cells.Results After treatment with different concentrations(25,50,100μmol/L)of Bicalutamide,the proliferation ability of MDA-MB-231 cells significantly reduced,and there was a time-and dose-dependent relationship.Bicalutamide could significantly increase apoptosis of the MDA-MB-231 cells and induced the cycle arrest at G0/G1 phase(P<0.05).Conclusion Bicalutamide can inhibit the proliferation of human triple negative breast cancer cells and potential molecular mechanism refers to cell apoptosis.
