GPRC5A、SOCS3和STAT3在结直肠癌组织中的表达及相关性分析 被引量:6

Expression and correlation of GPRC5A,SOCS3 and STAT3 in colorectal cancer tissues

摘要 目的探讨G蛋白偶联受体C家族5A(G protein-coupled receptor,family C,group 5,member A,GPRC5A)、信号转导和转录激活因子3 (signal transducer and activator of transcription 3,STAT3)和细胞因子信号转导抑制因子3(suppressor of cytokine signaling 3,SOCS3)在结直肠癌组织中的表达及与临床病理特征的关系。方法选取76例结直肠癌组织和30例癌旁正常组织标本,采用免疫组织化学法检测组织中GPRC5A、SOCS3和STAT3的表达情况,并分析其与患者临床病理特征的关系,采用Spearman相关分析方法分析三者表达的相关性。结果在结直肠癌组织中,GPRC5A和STAT3的阳性表达率均高于癌旁正常组织(59.2%vs 33.3%,P=0.016;69.7%vs 30.0%,P=0.001),而SOCS3的阳性表达率则低于癌旁正常组织(38.2%vs 80.0%,P=0.001)。GPRC5A表达水平与结直肠癌患者的肿瘤分化程度、淋巴结转移和TNM分期有关(P<0.05),SOCS3和STAT3的表达水平与结直肠癌患者的分化程度、肿瘤浸润深度、淋巴结转移和TNM分期有关(P<0.05)。Spearman相关分析显示,SOCS3与STAT3和GPRC5A的表达均呈负相关(r=-0.308,P=0.007;r=-0.340,P=0.003),GPRC5A与STAT3表达呈正相关(r=0.327;P=0.004)。结论 GPRC5A在结直肠癌组织中高表达,可能对SOCS3的表达具有负调控作用,对STAT3的表达有正调控作用,三者可能与结直肠癌的发生发展密切相关。 Objective To explore the expression of G protein-coupled receptor,family C,group 5,member A(GPRC5 A),signal transducer and activator of transcription 3(STAT3)and suppressor of cytokine signaling 3(SOCS3)in colorectal cancer tissues and their relationsip with clinicopathological features. Methods Totals of 76 colorectal cancer tissues and 30 adjacent normal tissues were enrolled. Immunohistochemistry was used to detect the expressions of GPRC5 A,SOCS3 and STAT3 in different tissues,and the relation ship between the expression and clinicopathological features and their correlation were analyzed by spearman correlation analysis. Results The positive expression rates of GPRC5 A and STAT3 in colorectal cancer tissues were significantly higher than those in adjacent normal tissues(59.2% vs 33.3%,P=0.016;69.7% vs 30.0%,P=0.001),but the positive expression rate of SOCS3 was significantly lower than that in adjacent normal tissues(38.2% vs 80.0%,P=0.001). The expression of GPRC5 A was significantly associated with pathological differentiation,lymph node metastasis,and TNM staging of colorectal cancer(P <0.05). The expression of SOCS3 and STAT3 were significantly associated with pathological differentiation,depth of invasion,lymph node metastasis,and TNM staging of colorectal cancer(P <0.05). The Spearman correlation analysis showed that SOCS3 protein expression were negatively correlated with STAT3 expression and GPRC5 A expression(r =-0. 308,P = 0. 007;r =-0. 340,P = 0. 003),and GPRC5 A protein expression was positively correlated with STAT3(r=0.327,P=0.004). Conclusions GPRC5 A is highly expressed in colorectal cancer tissues and may negatively regulate the expression of SOCS3 and positively regulate the expression of STAT3. GPRC5 A,SOCS3 and STAT3 may be closely related to the occurrence and development of colorectal cancer.
