摘要
基质辅助激光解吸/电离质谱(MALDI)中所用的溶剂对晶体的形成和质谱的质量起着重要的作用。研究了以二羟基苯甲酸(DHB)的x,j-异构体为基质的固相和液相紫外吸收材料。采用三种不同的溶剂体系制备固态DHB晶体,其中乙腈/水二元体系(L1)或乙醇/水二元体系(L2)是制备具有均匀尺寸晶体的最佳溶剂。该结果对UV-MALDI基质在成像、蛋白质组学和复杂混合物分析等领域的应用具有指导意义。
The solvent used in matrix-assisted laser desorption/ionization(MALDI)mass spectrometry can play a significant part in crystal formation and the quality of the mass spectrum.Both solid-state and liquid phase ultraviolet absorbing matrices based around x,j-isomers of dihydroxybenzoic acid(DHB)were evaluated.For the generation of solid-state DHB crystals,three different solvent systems were used.The most optimal solvent system in terms of generation of uniform size crystals was acetonitrile^water(L1)or ethanol/water(L2)binary system.These findings have implications for use of UV-MALDI ma¬trices in diverse applications such as imaging,proteomics and in complex mixture analyses.
