补肾化痰方影响去势骨质疏松大鼠Leptin及β2-AR表达的实验研究

Experimental study of Bushen Huatan Recipe on Leptin and β2-AR expression in castration osteoprosis rats

[目的]探讨补肾化痰方影响瘦素(Leptin)及β2肾上腺素能受体(β2-AR)的表达水平来防治绝经后骨质疏松症的机制。[方法]将75只大鼠随机分为假手术组、模型组、补肾化痰组、利维爱组、阿托伐他汀组,采用双侧卵巢摘除建立去势骨质疏松模型,假手术组只切除卵巢周围同等质量的脂肪组织。使用微计算机断层扫描技术(Micro-CT)检测大鼠右侧股骨的骨密度(BMD),酶联免疫吸附(ELISA)检测大鼠血清Leptin、雌二醇(E2),全自动生化仪检测甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C),聚合酶链反应(PCR)检测骨组织中Leptin mRNA、β2-AR mRNA相对表达量。[结果]模型组相对于假手术组BMD、血清Leptin、E2水平明显降低;治疗后,补肾化痰组相对于模型组的BMD、血清Leptin、E2、HDL-C含量及骨组织Leptin mRNA表达水平明显提高,血清TC、TG、LDL-L含量及骨组织β2-AR mRNA表达水平明显降低,与利维爱组及阿托伐他汀组比较无明显差异。[结论]补肾化痰方可以有效提高绝经后骨质疏松大鼠的BMD,补充雌激素水平,通过调节Leptin水平来介导交感神经的活性,起到防治绝经后骨质疏松和改善脂代谢的作用。

[Objective] To investigate the mechanism of Bushen Huatan Recipe on the expression of Leptin and β2-AR to prevent and treat postmenopausal osteoporosis. [Methods] The 75 rats were randomly divided into sham operation group,model group,Bushen Huatan group,livial group and atorvastatin group. The bilateral ovariectomy was used to establish the castration osteoporosis model. The sham operation group only the same quality adipose tissue around the ovary was removed. The bone density(BMD)of the right femur was measured by Micro-CT. The serum leptin and estradiol(E2)were detected by ELISA. Automatic biochemical analyzer for detection of triglyceride(TG),total cholesterol(TC),high density lipoprotein cholesterol(HDL-C),low density lipoprotein cholesterol(LDL-C).PCR detected the relative expression of Leptin m RNA,β2 adrenergic receptor (β2-AR)m RNA in bone tissue. [Results] The levels of BMD,serum Leptin and E2 in the model group were significantly lower than those in the sham operation group. After treatment,the levels of BMD,serum Leptin,E2,HDL-C and Leptin m RNA in the bone marrow of the Bushen Huatan group were significantly increased compared with those of the model group. Serum TC,TG,LDL-L and bone tissue β2-AR m RNA expression levels were significantly decreased,and there was no significant difference between the Livial group and the atorvastatin group. [Conclusion] Bushen Huatan Recipe can effectively improve the bone mineral density of postmenopausal osteoporosis rats,supplement estrogen levels,mediate the activity of sympathetic nerve by regulating the level of Leptin,prevent postmenopausal osteoporosis and improve lipid metabolism.
