龙须菜超滤多糖GCpF1的离子层析分离及生物活性研究 被引量:1

摘要 研究了龙须菜超滤多糖GCpF1的离子层析分离以及分离获得组分的生物学活性。GCpF1经离子凝胶层析分离,获得5个组分,主要由低盐洗脱组分GCpF1-2和GCpF1-3组成。免疫活性研究显示,5个多糖组分都可以刺激小鼠脾淋巴细胞增殖和激活小鼠骨髓来源巨噬细胞BMMs(Bone marrow-derived macrophages),并呈现剂量依赖关系,表明具有良好的免疫调节作用。5个组分中GCpF1-1活性最好,也优于GCpF1,而其余4个组分较GCpF1的活性略有降低。体外抑制肿瘤细胞株增殖试验表明,龙须菜多糖在体外不具有显著的细胞毒作用,对肿瘤细胞株L1210直接杀伤作用微弱。 The purification and bioactivity of polysaccharide GCpF1 which was isolated from Gracilaria lemaneiformis by ion chromatography were studied.GCpF1 was isolated to five fractions by ion chromatography.GCpF1-2 and GCpF1-3 which were eluted by low concentration of NaCl were the main elution.The immunocompetence assay showed that the five fractions could stimulate proliferation of mouse spleen lymphocyte and activate bone marrow macrophages(BMMs)in a dose-dependent manner,indicating a good immunoregulation activity on all fractions.Compared with GCpF1 and other fractions,the fraction GCpF1-1 had the best immunoregulation activity,but others were slightly descending.The assay of inhibition tumor cell proliferation showed that the polysaccharides form Gracilaria lemaneiformis had low toxicity for cell and couldn’t significantly kill tumor cells L1210 in vitro.
