Caspase-3、Caspase-8在兔腰椎固定融合后相邻节段椎间盘组织中的表达

Expression of Caspase-3 and Caspase-8 in Adjacent Segmental Intervertebral Disc Tissue after Rabbit Lumbar Fusion

摘要 目的观察针刺兔腰椎间盘后相邻节段椎间盘组织的细胞凋亡情况及凋亡相关蛋白Caspase-8、Caspase-3的表达,分析二者与椎间盘退变的关系。方法12个月龄健康新西兰大白兔16只,随机分成两组,实验组为针刺椎间盘退变模型组;对照组为针刺椎体,但未伤及椎间盘组,通过链霉亲和素-生物素-过氧化物酶复合物(SABC)免疫组织化学染色法及原位DNA片断末端标记法(TUNEL)检测细胞凋亡率及Caspase-8、Caspase-3表达。结果实验组可见细胞质空泡化,出现细胞团或多核巨细胞,凋亡细胞阳性率为51.3%;对照组可见髓核细胞多数呈孤立地存在于细胞陷窝内,细胞凋亡率为39.1%。实验组兔相邻节段椎间盘组织Caspase-8、Caspase-3蛋白表达阳性率分别为52.7%和47.8%;对照组兔Caspase-8、Caspase-3蛋白表达阳性率分别为35.9%和34.6%,实验组均高于对照组(P均<0.05)。结论针刺兔腰椎间盘引起退变后,其相邻椎间盘组织细胞凋亡率增高,Caspase-8、Caspase-3高表达说明细胞凋亡参与退变发生。 Objective To observe the apoptotic rate and the expression of apoptosis-related proteins Caspase-8 and Caspase-3 in the adjacent segmental intervertebral disc after acupuncture rabbit lumbar intervertebral disc,and to analyze the relationship between them and the degeneration of intervertebral disc.Methods Sixteen healthy New Zealand white rabbits of 12 months old were randomly divided into two groups.The experimental group was acupuncture intervertebral disc degeneration model group,and the control group was acupuncture vertebral body without damaging the intervertebral disc.The apoptosis rate and the expression of Caspase-8 and Caspase-3 were detected by SABC and TUNEL.Results In the experimental group,cytoplasmic vacuolation occurred,and cell clusters or multinucleated giant cells appeared.The positive rate of apoptotic cells was 51.3%.However,in the control group,most of the nucleus pulposus cells were isolated in the cell lacuna,and the apoptotic rate was 39.1%.The positive rates of Caspase-8 and Caspase-3 protein expression in the adjacent segmental intervertebral discs of the experimental group were 52.7%and 47.8%,respectively.The positive rates of Caspase-8 and Caspase-3 protein expression in the control group were 35.9%and 34.6%,respectively.The experimental group was higher than the control group(P all<0.05),and the difference was statistically significant.Conclusion After acupuncture degeneration of rabbit lumbar intervertebral disc,the apoptosis rate of adjacent intervertebral disc tissue is increased.The high expression of Caspase-8 and Caspase-3 indicates that apoptosis participates in degeneration.
