摘要 目的探讨阿司匹林(aspirin,ASA)对人结肠癌SW480细胞增殖、迁移和侵袭能力的影响。方法采用人结肠癌SW480细胞株,将0、1、2和4 mmol·L-1的ASA与细胞共培养作用24、48和72 h,通过CCK8实验观察ASA对SW480细胞增殖能力的影响;通过划痕实验、Transwell实验研究ASA对SW480细胞迁移、侵袭能力的影响;通过Western blot检测凋亡相关蛋白BAX、Bcl-2和肿瘤侵袭、迁移相关蛋白FAK、MMP9的表达情况。结果在同一时间下,与对照组相比,随着ASA作用浓度的增加SW480细胞的存活率降低(P<0.05);在同一浓度下,与对照组相比,随着ASA作用时间的增加,SW480细胞的存活率降低(P<0.05)。与对照组(0 mmol·L-1)相比,ASA能抑制结肠癌细胞增殖,降低Bcl2表达,提高Bax表达(P均<0.05);对SW480细胞迁移能力产生抑制(P<0.05),并下调迁移和侵袭相关蛋白FAK和MMP9的表达(P均<0.05)。结论ASA对结肠癌SW480细胞的迁移和侵袭能力有抑制作用,其抑制作用与FAK和MMP9蛋白表达下降有关。 Objective To investigate the effects of aspirin(ASA)on the proliferation,migration and invasion of human colon cancer SW480 cells.Methods Human colon cancer cell line SW480 was co-cultured with 0,1,2 and 4 mmol·L^-1 aspirin at different time points for 24,48 h and at 72 h,the effects of the drug on the proliferation of colon cancer SW480 cells were observed by CCK8 experiment.The effects of aspirin on the migration and invasion ability of SW480 cells were studied by scratch test,transwell cell migration and invasion experiments.The expression of apoptosis-related proteins Bax,Bcl-2,tumor invasion and migration related proteins FAK and MMP9 were detected by Western blot.Results At the same time,compared with the control group,the survival rate of SW480 cells decreased with the increase of aspirin concentration(P<0.05).At the same concentration,compared with the control group,with the increase of aspirin treatment time,the survival rate of SW480 cells decreased(P<0.05).Compared with the control group(0 mmol·L^-1),aspirin could inhibit the proliferation of colon cancer cells,reduce the expression of BCL2,increase the expression of Bax(P all<0.05),inhibit the migration of SW480 cells(P<0.05),and down regulate the expression of FAK and MMP9(P all<0.05).Conclusion Aspirin can inhibit the proliferation of colon cancer SW480 cells.Aspirin can induce apoptosis and promote the expression of BAX.Aspirin can inhibit the migration and invasion of colon cancer SW480 cells.Its inhibitory effect is related to the expression of FAK and MMP9 proteins in SW480 cells.
