摘要 目的通过体外实验研究成纤维细胞激活蛋白(Fibroblast activation protein,FAP)对卵巢癌细胞增殖和凋亡的影响。方法体外培养人类卵巢癌细胞系0VCAR3,用不同浓度的FAP(0、0.0625、0.125、0.25、0.5及1μg/mL)处理24 h或48 h后,通过MTT法检测FAP对卵巢癌细胞增殖的影响。0VCAR3用不同浓度的FAP(0、0.0625、0.25及1μg/mL)处理48 h后,通过流式细胞术检测FAP对OVCAR3细胞凋亡的影响。结果MTT法示FAP可促进卵巢癌细胞OVCAR3增殖,其作用呈时间、剂量依赖性(P<0.05);流式细胞术示FAP对卵巢癌细胞系OVCAR3的细胞凋亡率并无明显影响(P>0.05)。结论FAP可促进卵巢癌细胞系OVCAR3的增殖,对其凋亡水平无明显影响。 Objective To explore the effect of FAP on proliferation and apoptosis of ovarian cancer cell through in vitro experiment.Methods Human ovarian cancer cell line OVCAR3 was cultured in vitro and treated with FAP at different concentrations(0,0.0625,0.125,0.25,0.5 and 1μg/mL)for 24 or 48 hours.The effect of FAP on ovarian cancer cell proliferation was detected by MTT assay after treatment.After OVCAR3 was treated with FAP at different concentrations(0,0.0625,0.25 and 1μg/mL)for 48 hours,the effect of FAP on OVCAR3 apoptosis was detected by flow cytometry.Results It was shown by the MTT assay that FAP could promote OVCAR3 proliferation in a time-and dose-dependent manner(P<0.05).It was shown by the flow cytometry that FAP had no significant effect on the apoptosis rate of ovarian cancer cell line OVCAR3(P>0.05).Conclusion FAP can promote the proliferation of ovarian cancer cell line OVCAR3 but has no significant effect on its apoptosis level.
