沙葱黄酮对乌鳢血淋巴细胞活性、抗氧化能力及基因相对表达量的影响 被引量:6

Effects of flavonoids from Allium mongolicum Regel on the activity, antioxidant capacity and relative expression of blood lymphocyte of Channa argus

摘要 试验在体外环境下,研究沙葱黄酮对乌鳢血淋巴细胞活性、抗氧化能力及基因相对表达量的影响。试验在完全培养基中添加不同浓度的沙葱黄酮(0、25、50、100、200μg/ml)培养乌鳢血淋巴细胞,测定细胞活性;总超氧化物歧化酶(T-SOD)、过氧化氢酶(CAT)、总抗氧化能力(T-AOC)、谷胱甘肽过氧化物酶(GSH-Px)、丙二醛(MDA)及一氧化氮(NO)等抗氧化指标;以β-actin为内参基因,用实时荧光定量PCR法测定热休克蛋白70(HSP70)、热休克蛋白90(HSP90)、核因子κB抑制蛋白(IκBα)、糖皮质激素受体(GR)、白细胞介素-1(IL-1)、肿瘤坏死因子-α(TNF-α)、白细胞介素-8(IL-8)、核因子κB(NF-κB p65)基因相对表达量变化。结果显示:沙葱黄酮能显著提高乌鳢血淋巴细胞活性(P<0.05),提高抗氧化能力,上调HSP70、HSP90、IκBα、GR基因的表达,下调IL-1、TNF-α、IL-8、NF-κB p65基因的表达。试验结果表明沙葱黄酮能够有效提高乌鳢血淋巴细胞活性、抗氧化能力以及炎症相关基因的表达。 In this experiment,the effects of flavo⁃noids from Allium mongolicum Regel on the ac⁃tivity,antioxidant capacity and relative gene ex⁃pression of blood lymphocyte of Channa argus were studied under in vitro conditions.The ex⁃periment was carried out by adding different con⁃centrations of flavonoids from Allium mongoli⁃cum Regel(0,25,50,100,200μg/ml)to cul⁃ture medium,and measuring the cell activity and antioxidant parameters such as total antioxidant ca⁃pacity(T-AOC),superoxide dismutase(T-SOD),catalase(CAT),glutathione peroxidase(GSH-Px),malondialdehyde(MDA)and nitric oxide(NO);β-actin as an internal reference gene,measuring the relative expression of heat shock protein 70(HSP70),heat shock protein 90(HSP90),nuclear factor-κB inhibitor protein(IκBα),glucocorticoid receptor(GR),interleukin-1(IL-1),tumor necrosis factor-α(TNF-α),interleukin-8(IL-8),nuclear factor-κB(NF-κB p65)was determined by real-time quanti⁃tative PCR.The results showed that flavonoids from Allium mongolicum Regel could significantly in⁃crease the activity of blood lymphocyte of Channa argus(P<0.05),increase the antioxidant capacity,up-regulate the expression of HSP70,HSP90,IκBα,GR genes,and down-regulate the expression of IL-1,TNF-α,IL-8,NF-κB p65 genes.The results showed that flavonoids from Allium mongolicum Regel could effectively increase the activity of blood lymphocyte ofChanna argus,anti-oxidation abili⁃ty and expression of inflammation-related genes.
