摘要
目的探讨肿瘤坏死样因子凋亡微弱诱导剂(TWEAK)基因多态性与老年急性心肌梗死(AMI)的关系。方法回顾性分析2016年7月至2018年3月收治的、符合纳入标准的老年AMI患者126例作为病例组,并选择同期健康体检者132例作为对照组。提取外周血白细胞DNA,采用酶联免疫吸附法测定TWEAK水平,聚合酶链反应-限制性片段长度多态性(PCR-RFLP)技术检测rs11552708、rs12937543及rs4968211的基因型和等位基因。结果两组位点rs12937543基因型频率分布差异具有统计学意义(χ~2=8.06,P<0.05);两组位点rs11552708及rs4968211基因型频率分布差异均无统计学意义(χ~2=0.01、0.07,均P>0.05);两组位点rs12937543中A、T等位基因频率分布差异具有统计学意义(χ~2=5.79,P<0.05);携带等位基因T患AMI的风险是携带等位基因A者的1.74倍;病例组携带AT+TT基因者血清TWEAK水平高于对照组,差异具有统计学意义(P<0.01),两组携带AA基因者血清TWEAK水平差异无统计学意义(P>0.05);病例组携带T基因患者血清TWEAK水平高于非T基因携带患者,差异具有统计学意义(P<0.05)。结论 TWEAK多态性位点与AMI有关,TWEAK基因多态性可能参与了老年AMI的发生发展过程。
Objective To investigate the association between TWEAK gene polymorphism and acute myocardial infarction(AMI) in elderly patients. Methods One hundred and twenty-six elderly patients with AMI(case group) admitted July 2016 to March 2018 were enrolled in the study and 132 healthy subjects served as control group. Genotypes and alleles of TWEAK gene rs11552708, rs12937543 and rs4968211 were detected by PCR-RFLP. Results There was significant difference in the frequency distribution of rs12937543 genotype between the two groups(χ~2=8.06, P =0.02), but no significant differences in the frequency distribution of rs11552708 and rs4968211 genotype between two groups(χ~2=0.01, 0.07, P >0.05). The frequency distribution of A and T alleles in the two groups of rs12937543 was statistically significant(χ~2=5.79, P =0.02). The risk of AMI in subjects with allele T was 1.74 times higher than that with allele A. The serum TWEAK levels in patients with AT+TT genotype were higher than those in control group(P<0.01).There was no significant difference in the serum TWEAK level between patients with A genotype and those with non-A genotype(P >0.05);while the serum TWEAK level in patients with T genotype was higher than that with non-T genotype(P<0.01). Conclusion TWEAK gene rs12937543 polymorphism is associated with AMI in elderly patients.
