针刺不同腧穴组方对SHR大鼠血压及血清CGRP、ET-1和AngⅡ水平的影响 被引量:14

摘要 目的通过针刺不同穴位组方,观察对自发性高血压(SHR)大鼠血压及血清活性因子的影响,以期找到最佳的腧穴配伍组方。方法将72只9周龄雄性SHR大鼠随机分为9组,每组8只,即模型组、人迎组、曲池组、足三里组、曲池+足三里组、人迎+足三里组、人迎+曲池组以及三穴配伍组。另设8只Wistar-Kyoto (WKY)大鼠为空白对照组,不予针刺干预。每7天测定1次血压值,比较各组的血压变化水平,14周龄后检测大鼠血清中血管紧张素Ⅱ(AngⅡ)、降钙素基因相关肽(CGRP)和内皮素-1(ET-1)含量。结果针刺1周后,人迎组、人迎+曲池组、人迎+足三里组和三穴配伍组血压较针刺前下降(P<0.05),较模型组下降(P<0.05);针刺4周后各针刺组血压较模型组下降(P<0.05,P<0.01),与三穴配伍组比较,差异有统计学意义(P<0.05,P<0.01)。针刺后,各针刺组大鼠血清中的CGRP、ET-1和AngⅡ含量与模型组比较,差异均有统计学意义(P<0.05,P<0.01);除人迎+曲池组和人迎+足三里组的CGRP外,各针刺组的CGRP、ET-1和AngⅡ含量与三穴配伍组比较,差异均有统计学意义(P<0.05)。结论针刺不同腧穴组方可以降低SHR大鼠血压水平,其中人迎、曲池、足三里的三穴配伍组较其他腧穴组的降压效果显著,其机制可能通过调节血清中的CGRP、ET-1和AngⅡ含量来发挥作用。 Objective To investigate the effect of acupuncture at different prescriptions of acupoints on SHR blood pressure and serum active factors to find an acupoint prescription with the best effect.Method Seventy-two nine-week-old male SHRs were randomized to nine groups,namely model,Renying,Quchi,Zusanli,Quchi plus Zusanli,Renying plus Zusanli,Renying plus Quchi and three-acupoint combination groups,eight rats each.A blank control group composed of eight Wistar-Kyoto(WKY) rats was not treated with acupuncture.Blood pressure was measured once every seven days and compared between the groups.Serum angiotensin Ⅱ(Ang Ⅱ),calcitonin gene related peptide(CGRP) and endothelin-1(ET-1) contents were measured in the rats after 14 weeks of age.Result After one week of acupuncture,blood pressure decreased in the Renying,Renying plus Quchi,Renying plus Zusanli and three-acupoint combination groups compare with before(P<0.05) and with the model group(P<0.05).After four weeks of acupuncture,blood pressure decreased in every acupuncture group compared with the model group(P<0.05,P<0.01) and had a statistically significant difference compared with the three-acupoint combination group(P<0.05,P<0.01).After acupuncture,there were statistically significant differences in serum CGRP,ET-1 and Ang Ⅱ contents between every acupuncture group and the model group(P<0.05,P<0.01),and between every acupuncture group and the three-acupoint combination group except CGRP in the Renying plus Quchi and Renying plus Zusanli groups(P<0.05).Conclusion Acupuncture at different prescriptions of acupoints can reduce SHR blood pressure.The hypotensive effect is more marked in the three-acupoint(Renying,Quchi and Zusanli) combination group than in the other acupoint groups.Its mechanism of action may be through regulating serum CGRP,ET-1 and Ang Ⅱ contents.
