摘要
目的探讨石斛多糖对自发性高血压大鼠(SHR)炎症因子和晚期糖基化终产物代谢的效果及其剂量。方法提取石斛多糖,将SHR随机分为对照组、低剂量组、中剂量组和高剂量组四组,对照组给开水,低剂量组、中剂量组和高剂量组分别给石斛多糖100、300、500mg/d,干预2个月,比较干预后各组肿瘤坏死因子(TNF-α),白细胞介素(IL)-6,IL-10,晚期糖基化终产物(AGE),可溶性AGE受体(sRAGE)和全长AGE受体(RAGE)mRNA水平。结果与对照组比较,低剂量组TNF-α水平比较,差异有统计意义(P<0.05),中剂量组TNF-α、IL-6和IL-10水平差异有统计意义(P<0.05),高剂量组TNF-α、IL-6和IL-10水平差异有统计意义(P<0.05)。低剂量组sRAGE水平差异有统计意义(P<0.05),中剂量组sRAGE和RAGE mRNA水平比较,差异有统计意义(P<0.05),高剂量组AGE、sRAGE和RAGE mRNA水平比较,差异有统计意义(P<0.05)。石斛多糖能够有效缓解SHR的炎症因子和晚期糖基化终产物代谢紊乱。其改善TNF-α、IL-6、IL-10、AGE、sRAGE和RAGE mRNA的作用有剂量效应关系,纠正SHR炎症因子功能紊乱的有效剂量为300mg/d;纠正SHR晚期糖基化终末产物及其受体功能紊乱的有效剂量为500mg/d。结论石斛多糖可有效缓解SHR的炎症因子功能紊乱,降低AGE水平,提高sRAGE水平,降低RAGE基因的活化,对缓解SHR的血管重塑有积极的作用。其药效有剂量效应关系,中等剂量可有效改善炎症症状,高剂量能改善AGE及其受体症状。
Objective To explore effects of dendrobium polysaccharides on metabolic changes of spontaneous hypertensive rats(SHR)inflammatory factors and advanced glycation end products metabolism and its relationship with its dosage.Methods Dendrobium polysaccharides was extracted,and SHR was randomly divided into the control group,low dose group,medium dose group and high dose group.The control group was given boiled water,and the lowdose group,the middle-dose group and the high-dose group were given dendrobium polysaccharide of 100,300,500mg/d,respectively.After 2 months of intervention,tumor necrosis factor(TNF-α),interleukin(IL)-6,IL-10,mRNA levels of advanced glycation end products(AGE),soluble receptor for AGE(sRAGE)and full-length of receptor for AGE(RAGE)were compared between two groups after intervention.Results Compared with the control group,the difference of TNF-αlevels in the low dose group was statistically significant(P<0.05).Compared with the control group,the levels of TNF-α,IL-6 and IL-10 in the middle dose group were statistically significant(P<0.05).Compared with the control group,the levels of TNF-α,IL-6 and IL-10 in the high-dose group were statistically significant(P<0.05).The difference of sRAGE levels in the low dose group was statistically significant(P<0.05).The difference of sRAGE and RAGE mRNA levels in the middle dose group was statistically significant(P<0.05).The difference of AGE,sRAGE and RAGE mRNA levels in the high dose group was statistically significant(P<0.05).Dendrobium polysaccharide can effectively alleviate the inflammatory factors of SHR and the metabolic disorders of advanced glycation end products.It has a dose-effect relationship between the effects of improving TNF-α,IL-6,IL-10,AGE,sRAGE and RAGE mRNA.The effective dose to correct dysfunction of SHR inflammatory factors was 300mg/d.The effective dose to correct dysfunction of advanced glycation end products and their receptors in SHR was 500 mg/d.Conclusion Dendrobium polysaccharide can effectively alleviate the dysfunction of inflammatory factors in SHR,reduce AGE level,increase sRAGE level,reduce the activation of RAGE gene,which has a positive effect on relieving vascular remodeling of SHR.Its efficacy has a dose-effect relationship.Medium dose can effectively improve the symptoms of inflammation and high dose can improve the symptoms of AGE and its receptors.
