摘要
目的:研究依达拉奉对急性脑梗死大鼠的保护作用及生长分化因子15(GDF-15)的影响。方法:以大脑中动脉线栓栓塞法复制急性脑梗死大鼠模型,造模成功后大鼠随机分为模型组、依达拉奉高、低剂量组,并以假手术组作为对照。造模后1 h给药,24 h后进行神经行为学评分,取完整大脑进行TTC染色及梗死体积测量。通过RT-PCR及蛋白印迹法检测脑组织中GDF-15、Smad2 mRNA及蛋白的表达水平,并通过HE染色观察急性脑梗死大鼠大脑的病变程度。结果:模型大鼠神经行为学评分明显升高,脑梗死体积明显升高,脑组织病变严重,脑组织中GDF-15、Smad2 mRNA及蛋白的表达水平明显上升。不同剂量依达拉奉干预后,神经行为学评分明显下降,脑梗死体积明显减少,大脑病变程度减轻,脑组织中GDF-15、Smad2 mRNA及蛋白的表达水平明显下降。结论:依达拉奉对急性脑梗死大鼠的保护作用机制可能与其调节脑组织中GDF-15、Smad2的表达有关。
Objective:To study the protective effect of edaravone on acute cerebral infarction and GDF-15 in rats.Methods:The rat model of acute cerebral infarction was replicated by the tethered cerebral artery tethered method.The rats were randomly divided into the model group,the high and low dose groups of edaravone,and the sham operation group as the control.After modeling,1 h was administrated,and neurological behavioral scores were assessed after 24 h.TTC was taken from intact brain and infarcted size was measured.The expression of GDF-15,Smad2 mRNA and protein in brain tissue was detected by RT-PCR and Western blot,and the degree of cerebral lesion in rats with acute cerebral infarction was observed by HE staining.Results:The neurobehavioral score of the model rats increased significantly,the size of cerebral infarction increased significantly,the brain tissue was serious,and the expression level of GDF-15,Smad2 mRNA and protein in the brain tissue was obviously increased.After different doses of edaravone,the score of neurobehavioral score decreased significantly,the size of cerebral infarction decreased significantly,the degree of brain lesion was reduced,and the expression level of GDF-15,Smad2 mRNA and protein in the brain tissue decreased significantly.Conclusion:The protective mechanism of edaravone on rats with acute cerebral infarction may be related to regulating the expression of GDF-15 and Smad2 in brain tissue.
