摘要
目的:研究E-cadherin介导参与IL-24抑制肺癌细胞增殖和侵袭迁移的机制。方法:选用肺癌细胞A549作为体外研究对象,在细胞中转染pcDNA3.1-IL-24,Realtime PCR和Western blot方法测定过表达效果,MTT测定增殖变化,Transwell小室测定侵袭和迁移变化,Western blot检测E-cadherin、MMP-2、MMP-9和Ki-67蛋白表达变化。将E-cadherin shRNA和pcDNA3.1-IL-24共转染至肺癌细胞中,按照上述MTT、Transwell小室和Western blot方法测定细胞增殖、侵袭迁移和E-cadherin、MMP-2、MMP-9和Ki-67蛋白表达变化。结果:转染pcDNA3.1-IL-24后的肺癌细胞中IL-24表达水平升高,细胞增殖、侵袭和迁移能力下降,细胞中MMP-2、MMP-9和Ki-67蛋白表达水平降低,E-cadherin蛋白表达水平升高。E-cadherin shRNA可以逆转pcDNA3.1-IL-24对肺癌细胞增殖、侵袭和迁移的抑制作用及MMP-2、MMP-9和Ki-67蛋白表达的抑制作用。结论:E-cadherin参与IL-24抑制肺癌细胞增殖和侵袭迁移过程。
Objective:To study the mechanism of E-cadherin mediating IL-24 inhibiting proliferation and invasion and migration of lung cancer cells.Methods:Lung cancer cell A549 was selected as the object of study in vitro,pcDNA3.1-IL-24 was transfected into cells,Realtime PCR and Western blot were used to determine the overexpression effect,MTT was used to measure proliferation,Transwell chamber was used to measure invasion and migration,Western blot was used to detect the expression of E-cadherin,MMP-2,MMP-9 and Ki-67.E-cadherin shRNA and pcDNA3.1-IL-24 were co-transfected into lung cancer cells,the cell proliferation,invasion and migration,and the expression of E-cadherin,MMP-2,MMP-9 and Ki-67 were measured by MTT,Transwell chamber and Western blot.Results:The expression of IL-24 in lung cancer cells transfected with pcDNA3.1-IL-24 increased,the ability of cell proliferation,invasion and migration decreased,the expression levels of MMP-2,MMP-9 and Ki-67 in cells decreased,the expression of E-cadherin protein increased.E-cadherin shRNA could reverse the inhibitory effect of pcDNA3.1-IL-24 on proliferation,invasion and migration of lung cancer cells and the expression of MMP-2,MMP-9 and Ki-67 proteins.Conclusion:E-cadherin is involved in IL-24 inhibiting the proliferation and invasion and migration of lung cancer cells.
