摘要
目前在哺乳动物中已经发现了7个肿瘤坏死因子受体相关因子(TRAFs),主要参与肿瘤坏死因子受体超家族(TNFR)、Toll样/白细胞介素-1受体(TIR)、维甲酸诱导基因Ⅰ样受体(RLRs)的信号转导。TRAF3是TRAF家族中功能最为多样的成员之一。它能够正性调节Ⅰ型干扰素(IRFs)的产生,负性调节经典NF-κB、非经典NF-κB和丝裂原激活的蛋白激酶(MAPK)信号的激活。研究TRAF3在免疫信号通路中的作用、调控机制以及相关疾病具有重要的意义。
The mammalian tumor necrosis factor receptor associated factors(TRAFs)consists of seven members that are mainly involved in the signal translation of tumor necrosis factor receptor superfamily(TNFR),toll-like/interleukin-1 receptor(TIR)and retinoic acid induced gene-1 like receptor(RLRs).TRAF3 is a less characterized member in the TRAFs family.It can upregulate the production of typeⅠIFN,and negatively regulate signals of canonical NF-κB,non-canonical NF-κB and MAPK pathways.In this review,we focus on the role of TRAF3 in immune responses and the regulation mechanisms,as well as TRAF3 related diseases.
