摘要
目的探讨缺血再灌注损伤对大鼠海马组织N-甲基-D-天门冬氨酸受体(NMDAR)表达的影响。方法将24只SD大鼠按随机数字表法分成假手术组、对照组、模型组,每组8只,应用颈总动脉血管夹闭阻断法制作脑缺血再灌注损伤大鼠模型。应用HE染色法观察大鼠海马组织的损伤情况,采用免疫组织化学SABC法检测大鼠海马组织中NMDAR1的表达水平,并运用RT-PCR技术检测大鼠脑神经元的NMDAR1mRNA的变化。结果模型组海马组织缺血损伤使细胞溶解较假手术组严重;模型组大鼠海马组织中NMDAR1表达水平和NMDAR1mRNA含量较假手术组明显增多(P<0.01)。结论缺血再灌注通过引起兴奋性氨基酸的增多,相应地引起其NMDAR1表达增强。
Objective To investigate the effect of cerebral ischemical reperfusion injury on N-methyl-D-aspartate receptor(NMDAR)expression in rat hippocampus.Methods Twenty-four SD rats were randomly divided into sham operation group,control group and ischemic reperfusion group,with 8 rats in each group.The ischemic reperfusion was established by bilateral carotid artery occlusion.Hippocampal damage was observed by H&E staining.The expression of NMDAR1 in hippocampus was detected by immunohistochemical staining and RT-PCR.Results Hippocampal damage-caused cytolysis in ischemic reperfusion group was more serious than that in sham operation group.The expression of NMDAR1 mRNA and protein in ischemic reperfusion group was higher than that in sham operation group(P<0.01).Conclusion Cerebral ischemic reperfusion induces excitatory amino acid release and accordingly promotes the expression of NMDAR1 in hippocampus.
