电针对TBI后认知功能障碍大鼠MMP-2、MMP-9的调节作用

Moderating effect of electroacupuncture on MMP-2 and MMP-9 in rats with cognitive impairment after TBI

摘要 目的研究电针治疗TBI后认知功能障碍作用机制,为电针治疗TBI后认知障碍的临床应用提供科学证据。方法健康清洁SD雄性大鼠80只,经筛选、造模后共3组:空白组10只、模型组17只和电针组17只。选择百会、人中、内关、后三里、绝骨电针治疗,5次1疗程,共治2疗程。进行Morris水迷宫实验;血清中基质金属蛋白酶2(matrixmetalloproteinase-2、MMP-2)和基质金属蛋白酶9(matrix metalloproteinase-9,MMP-9)的测定。结果电针组大鼠水迷宫潜伏期时间和空间探索能力上调(P<0.01),血清中MMP-2、MMP-9含量下调(P<0.01)。结论电针能够通过调节血清中MMP-2、MMP-9的含量改善TBI后认知功能障碍大鼠的认知能力。 Objective To explore the mechanism of electro-acupuncture in the treatment of cognitive impairment after TBI, and to provide scientific evidence for the clinical application of electro-acupuncture in the treatment of cognitive impairment after TBI. Methods Eighty healthy and clean SD male rats were selected and modeled in three groups: 10 in the normal control group, 17 in the model control group and 17 in the electro-acupuncture treatment group. Baihui, Renzhong, Neiguan, Housanli and Juegu were treated with electro-acupuncture for 5 times and 1 course of treatment for 2 courses. Morris water maze test was carried out. Matrix metalloproteinase-2(MMP-2) and matrix metalloproteinase-9(MMP-9) in serum were measured. Results Electro-acupuncture could improve the latency time and space exploration ability of water maze in rats with cognitive impairment after TBI(P< 0.01), and decrease the levels of MMP-2 and MMP-9 in serum.Conclusion EA can improve the cognitive ability of rats with cognitive impairment after TBI by regulating the contents of MMP-2 and MMP-9 in serum.
