SPF级SD大鼠血液学指标95%参考区间的建立及性别差异探讨 被引量:4

摘要 目的建立8周龄SPF级SD大鼠血液学指标95%参考区间,并探讨性别因素的影响。方法在严格质量控制的条件下,选取本实验室近3年30d喂养试验终点阴性对照组SD大鼠,雌雄各160只,采用SIEMENS ADVIA 2120血液分析仪对其血液学指标进行检测,并制定95%参考区间。结果8周龄SPF级SD雌/雄大鼠血液学指标95%参考区间分别为:白细胞总数(WBC,×10^9/L):4.82~10.96/5.22~12.94;红细胞总数(RBC,×10^12/L):6.16~7.72/6.12~7.75;血红蛋白(HGB,g/L):111.40~143.46/114.00~143.00;血小板总数(PLT,×109/L):800.94~1456.33/862.21~1415.82;中性粒细胞百分比(NEUT%):4.94~24.58/6.78~24.94;淋巴细胞百分比(LYMPH%):69.83~91.48/69.59~89.73;单核细胞百分比(MONO%):0.96~4.12/1.20~4.80;嗜酸细胞百分比(EOS%):0.60~2.60/0.40~1.90;嗜碱细胞百分比(BASO%):0.10~0.30/0.10~0.30。WBC、NEUT%、MONO%雄鼠均高于雌鼠(t值分别为6.798、2.046、2.294,P值均<0.05),EOS%雄鼠低于雌鼠(t=-7.188,P<0.01),RBC、HGB、PLT、LYMPH%和BASO%雌雄之间无统计学差异(t值分别为-0.146、0.927、0.600、-1.662、0.779,P值均>0.05)。结论SD大鼠多项血液学指标存在雌雄差异,建立的95%参考区间可为保健食品毒理学安全性评价提供参考。 Objective To establish the reference ranges with 95%confidence levels of hematological indexes of 8 week-old SPF grade SD rats;to investigate the influence of genders.Methods Under strict quality control conditions,hematological indexes of 320 SD rats(160/gender)in control groups of 30 d-feeding experiments in past 3 years conducted by our laboratory were detected by SIEMENS ADVIA 2120 automatic hematology analyzer to establish the reference ranges with 95%confidence levels.Results The reference ranges with 95%confidence levels of hematological indexes of 8 week-old SPF grade female/male SD rats were as follows:WBC(×10^9/L):4.82-10.96/5.22-12.94;RBC(×10^12/L):6.16-7.72/6.12-7.75;HGB(g/L):111.40-143.46/114.00-143.00;PLT(×10^9/L):800.94-1456.33/862.21-1415.82;NEUT%:4.94-24.58/6.78-24.94;LYMPH%:69.83-91.48/69.59-89.73;MONO%:0.96-4.12/1.20-4.80;EOS%:0.60-2.60/0.40-1.90;BASO%:0.10-0.30/0.10-0.30.WBC,NEUT%and MONO%of male rats were higher than those of female rats(t=6.798,2.046,2.294;all P<0.05);EOS%of male rats was lower than that of female rats(t=-7.188,P<C0.01);indexes of RBC,HGB,PLT,LYMPH%and BASO%of SD rats showed no significant different between male and female rats(t=-0.146,0.927,0.600,-1.662,0.779,all P<0.05).Conclusion Multiple hematological indexes demonstrated significant differences between male and female rats.The established reference ranges with 95%confidence levels can provide reference for toxicological safety assessment of health foods.
