Optimization of Tissue Culture Conditions of Taraxacum kok-saghyz Rodin

摘要 以橡胶草为试验材料,探究种子的最佳表面消毒方法和不同植物生长调节剂对不定芽分化与生根的影响。结果表明:经过70%乙醇40 s和20%次氯酸钠10 min的表面消毒处理后,橡胶草种子的污染率最低,萌发率最高;当NAA浓度为0.1 mg/L,6-BA浓度为0.5 mg/L时,利于不定芽分化;当NAA浓度为0.3 mg/L时,利于橡胶草生根。不同外植体诱导不定芽分化的情况:根最快但易出现玻璃化现象;叶柄较快并有少量玻璃化发生;叶最慢且没有玻璃化现象出现。 Using Taraxacum kok-saghyz Rodin as the test material,the optimal surface disinfection method of seeds and the effects of different plant growth regulators on adventitious bud differentiation and rooting were investigated.The results showed that after 70%ethanol 40 s and 20%sodium hypochlorite 10 min surface disinfection,the Taraxacum kok-saghyz Rodin seeds had the lowest pollution rate and the highest germination rate;when the NAA concentration was 0.1 mg/L and the 6-BA concentration was 0.5 mg/L.It is conducive to adventitious bud differentiation;when the concentration of NAA is 0.3 mg/L,it is conducive to rooting of Taraxacum kok-saghyz Rodin.Different explants induced adventitious bud differentiation:The root was the fastest but prone to vitrification;The petiole was faster and a small amount of vitrification occurred;The leaf was the slowest and no vitrification occurred.
